摘  要:目的探讨糖皮质激素在RSV感染早期对干扰素信号通路的影响。方法RSV滴鼻建立RSV感染动物模型,腹腔注射地塞米松处理1 d和3 d后,肺泡灌洗液细胞计数及分类计数,ELISA检测肺泡灌洗液中IL-6、IL-8(KC)、IFN-γ、IFN-α、MMP-12水平以及肺匀浆中IFN-α水平,肺功能仪检测气道高反应。结果地塞米松不能抑制RSV感染引起的气道炎症和气道高反应,不能抑制炎症因子IL-6及KC的产生;地塞米松不影响肺匀浆中IFN-α水平但增加肺泡灌洗液中IFN-α水平,而对IFN-γ的水平无影响,且地塞米松抑制肺泡灌洗液中MMP-12的水平。结论地塞米松增加RSV感染早期IFN-α分泌水平,但不能抑制炎症反应。This study was performed to investigate the effect of dexamethasone on interferon signal pathway in the early stage of respiratory syncytical virus(RSV)infection.The RSV infected animal model was established by nose dropping of RSV,and dexamethasone was injected intraperitoneally.After 1 and 3 days of treatment,the cell count and differential cell count of BALF were measured.The levels of IL-6,IL-8(KC),IFN-γ,IFN-αand MMP-12 in BALF and the level of IFN-αin lung homogenate were detected by ELISA,and the hyperresponsiveness of airway was detected by pulmonary function instrument.Data showed that dexamethasone did not inhibit the airway inflammation or hyperresponsiveness caused by RSV infection,and did not inhibit the production of IL-6 or KC.But dexamethasone increased the level of IFN-αin BALF,while has no effect on IFN-αlevel in lung,or on the level of IFN-γ.More,dexamethasone inhibited the level of MMP-12 in BALF.Thus,dexamethasone may increase IFN-αlevel in the early stage of RSV infection,but can not inhibit the inflammatory response.
