摘 要:目的:探讨右美托咪定对老年小鼠剖腹探查术所致神经元凋亡的影响。方法:将40只16月龄的C57BL/6老年小鼠根据随机数字表随机分成4组:控制对照组(Con组)、右美托咪定组(Dex组)、手术组(Surgery组)、右美托咪定+手术组(Dex+Surgery组),每组10只。Con组腹腔注射等体积0.9%生理盐水;Dex组腹腔注射25μg/kg右美托咪定;Surgery组行剖腹探查术;Dex+Surgery组在行剖腹探查术前20分钟预先腹腔注射25μg/kg右美托咪定。手术结束6小时后,使用TUNEL试剂盒检测海马CA1区和DG区的神经元凋亡情况。用Western blot法检测海马凋亡相关蛋白表达情况(包括cleaved caspase-3、calpain 2、Bax、Bcl-2)。结果:与Con组相比,Surgery组老年小鼠海马CA1区和DG区TUNEL阳性细胞数明显增加(P<0.05),而与Surgery组相比,Dex+Surgery组老年小鼠海马CA1区和DG区TUNEL阳性细胞数明显降低(P<0.05)。Con组与Dex组海马CA1区和DG区TUNEL阳性细胞数无统计学差异(P>0.05)。与Con组相比,Surgery组cleaved caspase-3(P<0.05)、calpain 2(P<0.05)、Bax(P<0.05)表达明显升高,而Bcl-2(P<0.05)表达下降,与Surgery组相比,Dex+Surgery组cleaved caspase-3(P<0.05)、calpain 2(P<0.05)、Bax(P<0.05)表达明显下降,而Bcl-2(P<0.05)表达升高。Con组与Dex组海马cleaved caspase-3、calpain 2、Bax、Bcl-2无统计学差异(P>0.05)。结论:右美托咪定能抑制老年小鼠剖腹探查术所致神经元凋亡。Objective:To investigate the effect of dexmedetomidine on neuronal apoptosis induced by exploratory laparotomy in aged mice. Methods:Forty 16-month-old C57 BL/6 mice were randomly divided into four groups according to the table of random digit:Control group(Con group),Dexmedetomidine group(Dex group),Surgery group(Surgery group)and Dexmedetomidine+Surgery group(Dex+Surgery group),with 10 mice in each group. Con group received intraperitoneal injection of 0.9% normal saline with equal volume. Dex group was intraperitoneally injected with 25 μg/kg dexmedetomidine. Surgery group underwent exploratory laparotomy. Dex+Surgery group received intraperitoneal injection of 25 μg/kg dexmedetomidine 20 minutes before exploratory laparotomy. Six hours after the surgery,TUNEL kit was used to detect neuronal apoptosis in hippocampal CA1 and DG regions. The expression of apoptosis-related proteins(including cleaved caspase-3,calpain 2,Bax,and Bcl-2)in hippocampus was detected by Western blot. Results:Compared with Con group,the number of TUNEL-positive cells in hippocampal CA1 and DG regions of aged mice in Surgery group was significantly increased(P<0.05),which was significantly decreased in Dex+Surgery group(P<0.05)when compared with the Surgery group. There was no significant difference in the number of TUNEL positive cells in hippocampal CA1 and DG regions between Con group and Dex group(P>0.05). Compared with Con group,the expression of cleave caspase-3(P<0.05),calpain 2(P<0.05)and Bax(P<0.05)significantly increased,while the expression of Bcl-2(P<0.05)significantly decreased in Surgery group. Compared with Surgery group,the expression of cleave caspase-3(P<0.05),calpain 2(P<0.05)and Bax(P<0.05)significantly reduced,while the expression of Bcl-2(P<0.05)significantly increased in Dex+Surgery group. There is no significant difference in the expression of cleaved caspase-3,calpain 2,Bax and Bcl-2 between Con group and Dex group(P>0.05).
