摘 要:目的观察建中补肾消癥汤对肾间质纤维化中Wnt/β-catenin通路的影响。方法将120只小鼠随机分为假手术组,模型组,建中补肾消癥汤低剂量组、中剂量组、高剂量组,尿毒清组,采用单侧输尿管梗阻动物模型,观察14d、21d、28d后小鼠的形态变化,检测其血清肌酐、尿素氮水平,并检测梗阻侧肾组织中β-catenin、PAI-1、MMP7蛋白的表达。结果14、21、28d后模型组小鼠血清肌酐、尿素氮、β-catenin、PAI-1蛋白表达均高于假手术组(P<0.05),MMP7蛋白表达低于假手术组(P<0.05),经尿毒清组及各中药组治疗后小鼠血清肌酐、尿素氮、β-catenin、PAI-1蛋白表达较假手术组有所降低(P<0.05),MMP7蛋白表达有所升高(P<0.05),且建中补肾消癥汤高剂量组的疗效优于其他剂量及尿毒清组(P<0.05)。结论建中补肾消癥汤可能是通过调控Wnt/β-catenin通路以介导肾间质纤维化进程,延缓肾间质纤维化的发展以改善肾功能。Objective To observe the effect of the therapy of Jianzhong kidney-tonifying and Xiaozheng on Wnt/β-catenin pathway in renal interstitial fibrosis.Methods 120 mice were randomly divided into control group,model group,low dose group,middle dose group,high dose group,the urine poisonous clear group,using the animal model of unilateral ureteral obstruction to observe the morphology of mice,and detect the serum creatinine and BUN level,and testing theβ-catenin,PAI-1,MMP7 protein expression of the obstruction side kidney after 14,21,28 day.Results After 14,21,28 day,the serum creatinine and urea nitrogen,β-catenin,PAI-1 protein expression of model group mice were higher than in control group(P<0.05),MMP7 protein expression were lower than control group(P<0.05),the urine poisonous clear group and the Chinese medicine group after treatment serum creatinine and urea nitrogen in mice,β-catenin,PAI-1 protein expression has decreased in the control group(P<0.05),MMP7 protein expression was increased(P<0.05),and the curative effect of high dose group is superior to the other dose and uric poisonous clear group(P<0.05).Conclusion The therapy of Jianzhong kidney-tonifying and Xiaozheng may mediate the process of renal interstitial fibrosis by regulating Wnt/β-catenin pathway and delay the development of renal interstitial fibrosis to improve renal function.
