摘  要:目的:基于NF-κB/MAPK信号通路研究鱼藤素对过敏性哮喘(allergic asthma,AA)大鼠的作用机制。方法:雄性Wistar大鼠腹腔注射卵清蛋白(ovalbumin,OVA)致敏液[100μg OVA+1 mg Al(OH)3]加雾化OVA制备慢性哮喘大鼠模型,将模型制备成功的大鼠随机分为模型组、鱼藤素(低、中、高剂量)组(1 mg·kg^(-1)、2 mg·kg^(-1)、4 mg·kg^(-1)),每组8只,另取8只大鼠作为正常组,腹腔注射相应的药物7 d。检测大鼠的肺功能;ELISA法检测大鼠肺泡灌洗液中白细胞介素-1β(interleukin-1β,IL-1β)、IL-13水平;HE染色观察大鼠肺组织病理变化;Western Blot法检测大鼠肺组织p-NF-κB p65、NF-κB p65、p-p38 MAPK、p38 MAPK、p-ERK、ERK蛋白表达。结果:与模型组比较,鱼藤素低、中、高剂量组大鼠呼吸速率显著降低(P<0.05),吸气峰流量、呼气峰流量显著升高(P<0.05);大鼠肺泡灌洗液中IL-1β、IL-13水平显著降低(P<0.05);大鼠肺组织p-NF-κB p65/NF-κB p65、p-p38 MAPK/p38 MAPK、p-ERK/ERK水平显著降低(P<0.05);肺组织病理损伤有所改善。结论:鱼藤素可改善AA大鼠的肺功能及气道炎症反应,可能与抑制NF-κB/MAPK通路有关。Objective:Based on the NF-κB/MAPK pathway to study the mechanism of action of deguelin on allergic asthma(AA)rats.Methods:Male Wistar rats were injected intraperitoneally with aerosolized ovalbumin(OVA)sensitizer[100μg OVA+1 mg Al(OH)3]and aerosolized OVA to prepare chronic asthma models.Rats successfully modeled were randomly divided into a model group,deguelin low(1 mg·kg^(-1)),medium(2 mg·kg^(-1)),and high(4 mg·kg^(-1))dose groups,8 rats in each group,and another 8 rats as the normal group.The corresponding drugs were injected intraperitoneally for 7 days.The lung function of the rats was detected.The interleukin-1β(IL-1β)and interleukin-13(IL-13)levels in rat alveolar lavage fluid were detected by ELISA method,and expression of p-NF-κB p65,NF-κB p65,p-p38 MAPK,p38 MAPK,p-ERK,ERK protein in rat lung tissue were detected by Western Blot Method.Results:Compared with the model group,rats in the low,medium,and high dose groups of deguelin showed a significant decrease in respiratory rate(P<0.05),and a significant increase in peak inspiratory flow and peak expiratory flow(P<0.05).The levels of IL-1βand IL-13 in the lavage fluid were significantly reduced(P<0.05).And the levels of p-NF-κB p65/NF-κB p65,p-p38 MAPK/p38 MAPK,and p-ERK/ERK in rat lung tissue Significantly decreased(P<0.05).At the same time,the pathological damage of lung tissue was alleviated.Conclusion:Deguelin can improve lung function and airway inflammation in AA rats,which may be related to the inhibition of the NF-κB/MAPK signaling pathway.
