摘 要:[目的]从解毒酶系的角度研究了大菜粉蝶幼虫对坡柳皂苷的生理生化响应。[方法]用亚拒食剂量(AFC)的坡柳皂苷处理大菜粉蝶幼虫24、48 h后,测定试虫中肠内GSTs和CarE的活性,并采用透射电镜技术观察试虫中肠内质网的变化。[结果]药剂处理24 h时,羧酸酯酶和谷胱甘肽转移酶活性均显著高于对照,48 h时,谷胱甘肽转移酶活性仍显著高于对照,但羧酸酯酶活性反而显著低于对照,中肠内质网48 h时发生肿大膨胀。[结论]试虫体内解毒酶被激活是坡柳皂苷作用一段时间后拒食效果降低的原因之一,中肠内光面内质网的破坏可能导致了羧酸酯酶48 h活性的下降。为坡柳皂苷杀虫机理的阐明提供有益信息和线索。[Aims] This paper aims to study the physiological responses of the Pieris brassicae to Dononaea viscosa Saponins(DVS) by measuring detoxifying enzyme activity in 4 rd instar larvae. [Methods] Fed with AFC50, AFC30,AFC10 values of DVS in 24 and 48 h, the activities of glutathione-S transferase(GSTs) and carboxylesterase(CarEs)were measured, and transmission electron microscope was used to observe the midgut endoplasmic reticulum change in P. brassicae larvae. [Results] The results showed significantly higher activities of the GSTs and CarEs was found in 24 h,higher activities of the GSTs still went on but activies of CarE decreased in 48 h. Smooth endoplasmic reticulum was found expended. [Conclusions] The increased activity of the enzymes reduced the sensitivity of P. brassicae to DVS.The destruction of the smooth endoplasmic reticulum of the test insects is the cause of the decline of the CarE. The above results offer beneficial information for illustrating the insecticidal mechanism of the D. viscosa Saponins.
