摘 要:目的:探讨杜仲预处理对脑缺血再灌注损伤保护作用的最佳剂量。方法:采用随机数字表法将60只健康清洁级SD雄性大鼠分为6组,每组10只,分别是:假手术组、模型组、阳性药物组、杜仲低剂量组、杜仲中剂量组及杜仲高剂量组。除假手术组大鼠外其余各组均采用改良Zea-Longa线栓法制备局灶性脑缺血再灌注模型。比较各组大鼠脑缺血2 h再灌注24 h神经功能缺损评分、计算大脑含水量、Morris水迷宫比较学习记忆能力及观察脑组织病理学的差异。结果:(1)再灌注24 h,假手术组大鼠神经功能评分均为0分,其余各组大鼠均出现不同程度功能障碍,与模型组比较,阳性药物组、杜仲中剂量组、杜仲高剂量组大鼠神经功能评分均降低,且差异有统计学意义(P<0.05);(2)与模型组比较,阳性药物组、杜仲中剂量组及高剂量组大鼠脑含水量明显减轻,且差异有统计学意义(P<0.05);(3)造模前,模型组、阳性药物组、杜仲低剂量组、杜仲中剂量组、杜仲高剂量组大鼠与假手术组相比较,学习及记忆功能差异无统计学意义(P>0.05);造模后,阳性药物组、杜仲中剂量组、杜仲高剂量组与模型组相比较,学习记忆功能增强且差异有统计学意义(P<0.05);(4)通过HE染色观察,与模型组比较,阳性药物组、杜仲中剂量及高剂量组皮层结构紧密,神经元分布均匀。结论:杜仲可减轻局灶性脑缺血再灌注大鼠神经功能损伤、降低大脑含水量,有效改善大鼠造模后空间学习及记忆功能,对脑缺血再灌注损伤有一定的保护作用,且具有量效关系。Objective:To explore the optimal dose of Eucommia ulmoides pretreatment for cerebral ischemia-reperfusion injury(CIRI).Methods:60 male SD rats were randomly divided into the sham operation group,the model group,the positive drug group,the Eucommia ulmoides groups of low-dose,medium-dose and high-dose,with 10 rats in each group.The CIRI model was established by the modified Zea-Longa thread thrombus method.After 2 hours of cerebral ischemia and 24 hours of perfusion,the scores of neurological deficit,the calculation of brain water content,the ability of learning and memory and the pathological changes of brain tissue were compared in each group.Results:After 24 hours of reperfusion,the neurological function scores of the rats were all zero in the sham group,and the rats showed different degrees of dysfunction in the other groups;compared to those in the model group,the scores of neurological function were significantly decreased in the positive drug group and the Eucommia ulmoides groups of medium-dose and high-dose(P<0.05).Compared to that in the model group,the brain water content of the rats was significantly reduced in the positive drug group and the Eucommia ulmoides groups of medium-dose and high-dose(P<0.05).There were no statistical differences in learning and memory function among the model group,the positive drug group,the Eucommia ulmoides groups and the sham operation group before modeling(P>0.05);the learning and memory function was significantly higher in the positive drug group and the Eucommia ulmoides groups of medium-dose and high-dose than that of the model group after the intervention(P<0.05).Through HE staining observation,the cortical structure of the positive drug group and the Eucommia ulmoides groups of medium-dose and high-dose was tight and the distribution of neurons was uniform.Conclusion:Eucommia ulmoides can alleviate the injury of nerve function,decrease the water content of brain and effectively improve the spatial learning and memory function of the rats after modeling.It has a ce
