温阳化浊方对阿霉素肾病大鼠保护作用及TRPC6表达的影响

摘　　要：目的:探讨温阳化浊方对阿霉素肾病大鼠的治疗作用及其可能机制。方法:60只SPF级雄性SD大鼠随机分为对照组,模型组,泼尼松组,温阳化浊方高、中、低剂量组,每组10只。模型组及治疗组大鼠注射阿霉素进行造模。分别于给药后的2、4、8周检测24 h尿蛋白定量、血清甲状腺激素水平,光镜观察肾脏病理改变,免疫组化、激光共聚焦显微镜下及Western Blot法检测大鼠肾脏组织TRPC6蛋白表达。结果:(1)与模型组比较,各治疗组大鼠尿蛋白显著减少(P<0.05),血清T4升高,TSH水平降低(P<0.05);(2)光镜下各治疗组病理损害均有不同程度改善;(3)免疫组化、激光共聚焦显微镜及Western Blot检测显示:经过干预治疗后,各治疗组均能不同程度下调肾脏组织TRPC6蛋白的表达(P<0.05)。结论:温阳化浊方可明显减少尿蛋白量、改善甲状腺功能减退的状态、有效减轻肾脏病理损伤,其机制可能与下调TRPC6表达有关。Objective:To explore the therapeutic effect of Wenyang Huazhuo Formula(WHF)on Adriamycin-induced nephropathy in rats and its possible mechanism.Methods:60 male SD rats of SPF grade were randomly divided into the control group,the model group,the Prednisone group,the WHF groups of high-dose,medium-dose and low-dose,with 10 rats in each group.The model of nephrotic syndrome(NS)was established by the injection of Adriamycin.The levels of 24 h urine protein and serum thyroid hormones were measured at 2 w,4 w and 8w after the administration.The pathological changes of the rat renal tissues were observed under light microscope.Immunohistochemistry,laser confocal scanning microscope and western blot method were used to detect the protein expression of TRPC6 in rat renal tissues.Results:Compared to that in the model group,24 h urinary protein was significantly decreased(P<0.05),and the serum level of T4 was elevated and the serum level of TSH was lowered in each treatment group(P<0.05).The pathological damage was improved to different degrees in each treatment groups under light microscope.In terms of the tests revealed by immunohistochemistry,laser confocal scanning microscope and western blot method,the protein expression of TRPC6 was significantly down-regulated in different treatment groups(P<0.05).Conclusion:WHF could significantly reduce the amount of urine protein,improve the state of hypothyroidism and reduce renal pathological damage effectively,which mechanism may be related to the down-regulation of TRPC6 expression.
