摘  要:试验旨在探究PAX3蛋白的结构和功能,对后期研究其参与的生理机制提供理论依据。利用生物信息学技术对牛PAX3基因结构、蛋白保守结构域、信号肽剪切位点、理化性质、亲疏水性和亚细胞定位进行分析,并对其二级三级结构进行预测。结果显示,牛PAX3基因编码氨基酸在人、小鼠、大鼠、牛、猪、马、鸡7个物种中高度保守,且均含有配对结构域PAX、低复杂性区域和完全同源结构域HOX;牛PAX3蛋白等电点8.81,半衰期30 h,不稳定指数56.68,为亲水性蛋白,在细胞核分布的概率95.70%,二级结构中存在43个α螺旋,441个无规卷曲,不存在β折叠。综上所述,推测PAX3蛋白存在于细胞核中,通过与亲水性蛋白结合发挥其生物学功能。The purpose of the study was to explore the structure and function of PAX3 protein,and to provide theoretical basis for later studies on the physiological mechanisms involved in PAX3.Bioinformatics techniques were used to analyze the gene structure,protein conserved domain,signal peptide splice site,physicochemical properties,hydrophilicity and subcellular localization of bovine PAX3,and to predict its secondary and tertiary structure.The results showed that the amino acid of bovine PAX3 protein was highly conserved in human,mouse,rat,bovine,pig,horse and chicken,and all of them contained PAX domain,the low complexity domain and Hox domain.The isoelectric point of bovine PAX3 protein was 8.81,the half-life was 30 h,the instability index was 56.68,and it was hydrophilic protein.The probability of distribution in the nucleus was 95.70%,and there were 43αhelices and 441 random coils in the secondary structure.In summary,it is speculated that PAX3 protein exists in the nucleus and plays its biological function by binding with hydrophilic proteins.
