摘 要:目的以C57BL/6小鼠为动物模型,观察美洲大蠊Periplaneta americana胸腺素对小鼠毛发生长及生长周期的影响。方法将背部脱毛的C57BL/6小鼠分为PBS组(空白处理)、THY1组(给药浓度500μg·mL^(-1))、THY2组(给药浓度500μg·mL^(-1))、THY3组(给药浓度500μg·mL^(-1))和Tβ4组(给药浓度500μg·mL^(-1)),连续给药18 d,记录小鼠皮肤颜色变化时间、脱毛区毛发生长状况以及毛囊组织学特征和毛囊数目的变化。通过实时荧光定量PCR法检测胰岛素样生长因子-1(IGF-1)、β-连环蛋白(β-catenin)和基质金属蛋白酶-2(MMP-2)的表达水平。结果与PBS组相比,给药组小鼠脱毛后皮肤颜色由粉红变黑的时间更短,毛囊数量均明显更多,且给药组小鼠毛发生长更快。在给药第6天,THY1组和THY3组的β-catenin表达明显高于PBS组;给药第18天,THY2组的MMP-2表达明显高于PBS组,且THY1组的IGF-1表达明显高于PBS组。结论美洲大蠊胸腺素可以促进C57BL/6小鼠的毛发生长,可能通过提高β-catenin、MMP-2和IGF-1的表达来调控小鼠的毛囊发育与毛发生长。Objective To explore the effect of Periplaneta americana thymosin on hair growth and hair cycle of C57BL/6 mouse.Methods The C57BL/6 mice were divided into THY1 group,THY2 group,THY3 group and Tβ4 group with administration concentration of 500μg·mL^(-1),and continuously administered for 18 days.Photographs were taken daily to record the changes in skin color and hair growth status of mice with hair removal.The effect of P.americana thymosin on hair growth of mice was determined by the changes of hair follicle morphology and quantity.The expression levels of IGF-1,β-catenin and MMP-2 in the skin were detected by real-time fluorescence quantitative PCR(qPCR)on day 6,day 12 and day 18.Results Compared with the PBS group,the skin color of mice from the administration groups changed in a shorter time after hair removal,and the number of hair follicles was significantly higher,and the hair growth was faster.Compared with the PBS group,the expression levels ofβ-catenin in the THY1 and THY3 groups were significantly higher on day 6,while the expression of MMP-2 in the THY2 group and the expression of IGF-1 in the THY1 group were significantly higher on day 18.Conclusion P.americana thymosin can promote the hair growth of C57BL/6 mice,and the increased expression ofβ-catenin,MMP-2 and IGF-1 may be one of the mechanisms by which P.americana thymosin can regulate hair follicle development and hair growth.
