摘 要:为了明确椰心叶甲啮小蜂Tetrastichus brontispae响应种群退化的关键功能基因,通过对比其复壮和保种种群的转录组数据,对差异表达基因进行GO富集分析,并将差异基因在KEGG数据库中进行对比,获得显著性富集的通路注释信息。结果表明:相较于复壮种群,保种种群共有553个差异表达基因,其中206个基因上调表达,347个基因下调表达;这些差异基因显著富集在分子功能、细胞组分和生物过程三个大类中。分子功能中,富集到单氧酶活动的差异基因数量最多,占供试基因总数的28.57%;细胞膜富集到细胞组分中的差异基因最多,占供试基因总数的68%;而富集在生物过程中各项的差异基因都较少,仅有1~2个。差异基因主要在Hh信号通路、Hh信号通路-fly、胰腺分泌和神经活性配体受体相互作用4条通路中显著富集。椰心叶甲啮小蜂复壮和保种种群的差异基因涉及到蜂王浆主蛋白、嗅觉蛋白、细胞色素P450等,与椰心叶甲啮小蜂的个体发育、求偶交配以及产卵过程密切相关,可为进一步探究椰心叶甲啮小蜂种群退化的分子机制奠定理论基础。To identify the key functional genes in response to population degradation of Tetrastichus brontispae.GO enrichment analysis was performed on differentially expressed genes,and the differentially expressed genes were compared in the KEGG database to obtain significant enrichment pathway annotation information by comparing the transcriptome data of rejuvenated and conserved populations.Compared with the rejuvenated population,a total of 553 differentially expressed genes(DEGs)were found by analyzing the transcriptome data in the conserved population of T.brontispae,among which 206 genes were up-regulated and 347 genes were down-regulated.These DEGs were significantly enriched in three categories:molecular function,cell component and biological process.In molecular function,the most enriched DEGs was in the activity of monooxygenase,accounting for 28.57%of the total tested genes,and in cell component,the most enriched DEGs was in cell membrane,accounting for 68%of the total tested genes,and enriched in each term of biological process,there are only one or two differential genes.DEGs were significantly enriched in four pathways:Hh signaling pathway,Hh signaling pathway-fly,pancreatic secretion and neuroactive ligand receptor interaction.The differential genes between the two populations mainly involved major royal jelly protein,olfactory protein and cytochrome P450,which were closely related to the ontogeny,courtship and oviposition process of T.brontispae.DEGs of two populations were analyzed based on transcriptome information,which provided a theoretical basis for further exploring the molecular mechanism of population degradation of T.brontispae.
