摘 要:目的:采用HPLC法测定不同产地乌药叶中芦丁、金丝桃苷、异槲皮苷和槲皮苷的含量,构建其指纹图谱,并结合化学模式识别方法,评价乌药叶的质量。方法:采用Agilent HC-C_(18)色谱柱,以乙腈(A)和0.4%磷酸溶液(B)为流动相进行梯度洗脱,流速为1.0 mL/min,柱温为30℃,检测波长为343 nm,进样量为10μL。结果:芦丁、金丝桃苷、异槲皮苷、槲皮苷分别在0.0967~0.7733、0.0967~1.9333、0.1083~2.1667、0.0933~37.3333μg范围内线性关系良好。聚类分析将16批乌药叶分为4类;主成分分析筛选出2个主成分,累计方差贡献率为89.002%,且碎石图中有2个较陡坡度主成分。结论:湖南不同产地乌药叶中芦丁、金丝桃苷、异槲皮苷、槲皮苷的含量有所差异,可能与其生长环境和采收时间有关;其中异槲皮苷、槲皮苷对主成分有着较大影响,可为乌药叶的多成分含量测定提供依据。Objective:To determine the contents of rutin,hyperoside,isoquercitrin and quercitrin in Wuyaoye(Linderae Folium)from different habitats by HPLC,to establish their fingerprints,and to evaluate the quality with chemical pattern recognition methods.Methods:Agilent HC-C_(18) column was used for gradient elution with acetonitrile(A)and 0.4%phosphoric acid solution(B)as the mobile phase.The flow rate was 1.0 mL/min,the column temperature was 30℃,the detection wavelength was 343 nm and the injection volume was 10μL.Results:Rutin,hyperoside,isoquercitrin,and quercitrin have good linear relationships in the range of 0.0967-0.7733,0.0967-1.9333,0.1083-2.1667 and 0.0933-37.3333μg,respectively.The cluster analysis divided 16 batches of Wuyaoye(Linderae Folium)into 4 categories;the principal component analysis screened out 2 principal components,the cumulative variance contribution rate was 89.002%,and there were 2 steeper slope principal components in the gravel map.Conclusion:The contents of rutin,hyperoside,isoquercitrin,and quercitrin in Wuyaoye(Linderae Folium)from different habitats in Hunan are different,which may be related to the growth environment and harvest time.Among them,isoquercitrin and quercitrin have a greater influence on it,which can provide a basis for the determination of the content of the multi-components of Wuyaoye(Linderae Folium).
