摘 要:细胞外基质(extracellular matrix, ECM)是一种由水合大分子蛋白质及多糖组成的复杂网络结构,构成组织的无细胞基质微环境。作为天然生物材料,ECM一直是组织工程器官/组织体外制备的理想载体支架来源。随着天然组织中ECM对细胞影响的发现,ECM调节细胞行为的生物化学作用越来越受到关注。该文系统概述了不同类型的ECM对细胞黏附、迁移、增殖和分化等细胞行为的影响,重点分析了ECM对细胞行为的调控作用机理,并阐述了基于模拟ECM的人工支架在器官/组织再生中的应用进展,为全面了解ECM对细胞行为的调控作用机制及其再生应用潜能提供建议与参考。Extracellular matrix(ECM) is a complex network structure composed of hydrated macromolecular proteins and polysaccharides, which constitutes the cell-free matrix microenvironment. As a natural biomaterial, ECM has always been an ideal source of biomaterials for the construction of the tissue engineering organ/tissue in vitro. With the discovery of the regulatory effect of ECM on cells in natural tissues, more and more attention has been paid to the biochemical effect of ECM on regulating cell behavior. In this article, the effects of ECM on cell adhesion, migration, proliferation and differentiation were systematically summarized, and the regulatory mechanisms of different ECM on cell behavior were analyzed. Furthermore, the application progress of artificial scaffolds based on ECM in tissue/organ regeneration was elaborated, which provided suggestions and reference for a comprehensive understanding of the regulatory mechanism of ECM on cell behavior and regeneration application.
