摘 要:为完善厚朴有效成分提取方法,采用正交试验设计,HPLC测定厚朴中厚朴酚与和厚朴酚含量的方法,研究了水浴辅助条件下利用超声提取厚朴中酚类物质最佳方案。结果表明,在色谱条件SinoChrom ODS-BP 5 um 4.6 mm×150 mm色谱柱,流动相为甲醇—水(78∶22),流速为1.0 ml/min,检测波长294 nm,柱温30℃,进样量20 ul,乙醇浓度90%,浸泡时间50 min,浸泡温度60℃,超声时间20 min条件下,酚类物质分离效果最优,提取率最高。In order to study the extraction of phenolic substances from Magnolia officinalis by means of ultrasound with the aid of a water bath,the best extraction scheme was selected.In this study,an orthogonal design was used to determine the content of magnolol and honokiol in Magnolia officinalis by HPLC.The results showed that under the chromatographic conditions,with SinoChrom ODS-BP 5 um 4.6 mm×150 mm chromatographic column,the mobile phase as methanol-water(78∶22),the flow rate at 1.0 ml/min,the detection wavelength at 294 nm,the column temperature at 30℃,the injection volume as 20 ul,the ethanol concentration 90%,the immersion time for 50 min,the immersion temperature at 60℃,and the ultrasonic time for 20 min,the separation effect of similar substances is the best and the extraction rate is the highest.
