摘 要:目的探讨大黄素对急性肺损伤模型大鼠的保护作用。方法将40只SD大鼠随机分为对照组、模型组及大黄素低、高剂量组,各10只。除对照组外,其余3组大鼠均尾静脉注射油酸(0.1mL/kg)以复制急性肺损伤模型,建模成功后,大黄素低、高剂量组大鼠分别给予10,20mg/kg大黄素灌胃,对照组和模型组予等体积0.9%氯化钠溶液灌胃,每天1次,连续14d。采用全自动血气分析仪检测大鼠动脉血pH、氧分压(PaO_(2))和氧合指数(PaO_(2)/FiO_(2)),测定大鼠肺组织湿干质量比(W/D);以苏木素-伊红(HE)染色,观察大鼠肺组织病理学变化;采用酶联免疫吸附(ELISA)法检测大鼠肺泡灌洗液中白细胞介素1β(IL-1β)、白细胞介素6(IL-6)和肿瘤坏死因子-α(TNF-α)水平;采用Westernblotting法检测大鼠肺组织中水通道蛋白4(AQP4)和核因子-κB(NF-κB)蛋白表达水平。结果与模型组比较,大黄素低、高剂量组大鼠动脉血pH,PaO_(2),PaO_(2)/FiO_(2)均显著升高;肺组织的W/D及其中AQP4蛋白和p-NF-κBp65蛋白表达水平,肺组织学评分及肺泡灌洗液中IL-1β,TNF-α,IL-6水平均显著降低(P<0.05)。结论大黄素对模型大鼠的急性肺损伤状态有一定改善作用,其机制可能与下调炎性细胞因子和AQP4表达水平及抑制NF-κB信号通路有关。Objective To investigate the protective effect of emodin on acute lung injury model rats.Methods A total of 40 SD rats were randomly divided into the control group,model group and low-dose and high-dose emodin groups,10 rats in each group.In addition to the rats in the control group,the rats in the other three groups were injected with oleic acid(0.1 m L/kg)through the tail vein to replicate the acute lung injury model.After successful modeling,the rats in the low-dose and high-dose emodin groups were given 10,20 mg/kg of emodin by gavage respectively,and the rats in the control group and the model group were given the equal volume of 0.9%Sodium Chloride Solution by gavage,once a day for 14 d.The arterial blood pH,partial pressure of oxygen(PaO_(2))and oxygenation index(PaO_(2)/FiO_(2))were measured by the automatic blood gas analyzer,and the wet-dry mass(W/D)ratio of the lung tissues of rats in each group was measured.Hematoxylin-eosin(HE)staining was used to observe the pathological changes of lung tissue of rats.The levels of interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)in the bronchoalveolar lavage fluid(BALF)of rats was detected by the enzyme-linked immunosorbent assay(ELISA)method.The expression level of aquaporin 4(AQP4)and nuclear factor-κB(NF-κB)protein in the lung tissue of rats were detected by Western blotting.Results Compared with those in the model group,the arterial blood pH,PaO_(2) and PaO_(2)/FiO_(2) of rats in the low-dose and high-dose emodin groups were significantly higher,the W/D ratio of lung tissue,the expression levels of AQP4 protein and p-NF-κBp65 protein,the lung histological score and the levels of IL-1β,TNF-αand IL-6 in the BALF of rats in the low-dose and high-dose emodin groups were significantly lower(P<0.05).Conclusion Emodin can improve the state of acute lung injury in model rats,and its mechanism may be related to down-regulating the expression of inflammatory cytokines and AQP4,and inhibiting the activity of NF-κB signaling pathway.
