高效液相色谱法同时测定马尾连及硬水黄连提取物中4种生物碱类成分含量

摘　　要：目的建立同时测定近缘植物马尾连和硬水黄连提取物中木兰花碱、药根碱、黄连碱、小檗碱4种生物碱类成分含量的高效液相色谱法。方法色谱柱为Zorbax Eclipse XDB-C_(18)柱(250mm×4.6mm,5μm),流动相为0.1%磷酸水溶液加0.3%十二烷基硫酸钠-乙腈(梯度洗脱),流速为1mL/min,检测波长为268nm,柱温为35℃,进样量为10μL。结果木兰花碱、药根碱、黄连碱、小檗碱质量浓度分别在2.158~86.300mg/L、5.695~227.800mg/L、3.815~152.600mg/L、4.488~179.500mg/L范围内与峰面积线性关系良好;精密度、稳定性、重复性试验的RSD均小于3%(n=6);马尾连提取物中木兰花碱、药根碱、黄连碱、小檗碱的平均加样回收率分别为98.94%,99.27%,99.70%,99.11%,RSD分别为0.96%,0.80%,1.19%,1.52%(n=6),含量分别为0.797,2.532,1.591,1.747mg/g;硬水黄连提取物中黄连碱和小檗碱的平均加样回收率分别为98.48%和98.90%,RSD分别为1.13%和1.85%(n=6),含量分别为1.960,2.401mg/g。结论该方法简便快捷,准确度高,重复性和稳定性均较好,可用于木兰花碱、药根碱、黄连碱、小檗碱4种生物碱类成分的含量测定。Objective To establish a high-performance liquid chromatography(HPLC)method for the simultaneous determination of the content of four alkaloids(magnoflorine,jatrorrhizine,coptisine and berberine)in the extract of Radix et Rhizoma Thalictri and Radix Thalictri Brveipis.Methods The chromatographic column was Zorbax Eclipse XDB-C_(18) column(250 mm×4.6 mm,5μm),the mobile phase was 0.1%phosphoric acid aqueous solution plus 0.3%sodium dodecyl sulfate-acetonitrile(gradient elution),the flow rate was 1 m L/min,the detection wavelength was 268 nm,the column temperature was 35℃,and the injection volume was 10μL.Results The linear ranges of magnoflorine,jatrorrhizine,coptisine and berberine were 2.158-86.300 mg/L,5.695-227.800 mg/L,3.815-152.600 mg/L and 4.488-179.500 mg/L.The RSDs of precision,stability and repeatability tests were less than 3%(n=6).The average recoveries of magnoflorine,jatrorrhizine,coptisine and berberine in the extract of Radix et Rhizoma Thalictri were 98.94%,99.27%,99.70%and 99.11%with RSDs of 0.96%,0.80%,1.19%and 1.52%(n=6),and their content was 0.797,2.532,1.591,1.747 mg/g,respectively.The average recoveries of coptisine and berberine in the extract of Radix Thalictri Brveipis were98.48%and 98.90%with RSDs of 1.13%and 1.85%(n=6),and their content was 1.960,2.401 mg/g,respectively.Conclusion The method is simple,rapid,accurate,reproducible and stable,which can be used for the content determination of four alkaloids(magnoflorine,jatrorrhizine,coptisine and berberine).
