一测多评法测定礞石滚痰丸中6种成分含量

摘　　要：目的建立同时测定礞石滚痰丸中6种成分含量的一测多评法。方法色谱柱为Thermo ODS Hypersil柱(250mm×4.6mm,5μm),流动相为甲醇-0.1%磷酸水溶液(梯度洗脱),流速为1.0mL/min,检测波长为254nm,柱温为30℃。以大黄素为内参物,建立大黄素与黄芩苷、芦荟大黄素、大黄酸、大黄酚、大黄素甲醚的相对校正因子。采用一测多评法测定礞石滚痰丸中6种成分的含量;同时以外标法进行验证试验。结果大黄素与黄芩苷、芦荟大黄素、大黄酸、大黄酚、大黄素甲醚的相对校正因子分别为0.26,1.47,0.11,2.13,0.34,在不同试验条件下相对校正因子重复性良好,6种成分含量的计算值与实测值无显著差异(P>0.05)。结论所建立的方法科学、简便、成本较低,可用于同时测定礞石滚痰丸中多种有效成分的含量。Objective To establish a quantitative analysis of multi-components by single-marker(QAMS)method for the simultaneous determination of six components in Mengshiguntan Pills.Methods The chromatographic column was Thermo ODS Hypersil column(250 mm×4.6 mm,5μm),the mobile phase was methanol-0.1%phosphoric acid solution(gradient elution),the flow rate was 1.0 m L/min,the detection wavelength was 254 nm,and the column temperature was 30℃.With emodin as the internal reference material,the relative correction factors of emodin and baicalin,aloe-emodin,rhein,chrysophanol and physcion were established.The contents of six components in Mengshiguntan Pills were determined by the QAMS method.At the same time,the external standard method(ESM)was used for the verification test.Results The relative correction factors of emodin and baicalin,aloe-emodin,rhein,chrysophanol and physcion were0.26,1.47,0.11,2.13 and 0.34,respectively.Under different experimental conditions,the reproducibility of the relative correction factors was good,and there was no significant difference between the calculated values and the measured values of the six components(P>0.05).Conclusion The established method is scientific,simple with low cost,which can be used for the simultaneous determination of multiple effective components in Mengshiguntan Pills.
