摘  要:目的研究去甲斑蝥素(norcantharidin, NCTD)诱导的超氧阴离子(superoxide anion)对人宫颈癌HeLa细胞毒性及作用机制。方法甲基噻唑蓝(MTT)法检测60μmol/L NCTD作用后对HeLa的细胞生长抑制作用,采用流式细胞术检测HeLa细胞凋亡率和细胞周期,蛋白印迹法检测细胞周期相关蛋白的表达。结果 60μmol/L NCTD作用24 h后能抑制HeLa细胞增殖和诱导HeLa发生细胞凋亡,下调cyclin B1的表达水平介导G2/M期周期阻滞。5 mmol/L超氧阴离子清除剂超氧化物歧化酶(SOD)作用后显著降低NCTD诱导的HeLa细胞生长抑制和细胞凋亡率,逆转NCTD诱导的cyclin B1细胞周期蛋白表达水平降低。结论 NCTD介导细胞内超氧阴离子的产生,调控HeLa细胞发生G2/M期周期阻滞。Objective To study the cytotoxicity and mechanisms of superoxide anioninduced by Norcantharidin(NCTD) in HeLa cells.Methods MTT assay was performed to assess the inhibition of 60 μmol/L NCTDin HeLa. The cell cycle alteration and apoptotic ratio were detected by flowcytometry. The expression levels of cell cycleprotein were examined by Western blotting.Results NCTD could inhibit HeLa cell proliferation and induced cell apoptosis;Western blotting showed that NCTD significantly induced G2/M cell cycle arrest associated with down-regulated cyclin B1expression.Besides, SOD(superoxide dismutase), the superoxide anion scavenger, extremely decreased the growth-inhibitory ratio and cell apoptosis in NCTD-treated HeLa cells. Moreover, NCTD-induced expressionof cyclin B1 were apparently reversed by 5 mmol/L SOD in HeLa cells.Conclusion Norcantharidin increased the production of superoxide anions to induce G2/Mcell cycle arrest in HeLa cell.
