摘 要:目的:探究中药冬虫夏草改善肝脏纤维化的调控机制。方法:基于转录组二代高通量测序(RNA-seq)的数据结果分析肝纤维化大鼠与正常大鼠的差异表达基因并找出差异基因的富集通路。大鼠随机分为对照组、模型组(四氯化碳诱导肝纤维化模型)、冬虫夏草(1 500 mg·kg^(-1)·d^(-1))组。经过6周的灌胃治疗,取大鼠的眼眶血用于肝功能测定,提取肝脏中RNA用于分子生物研究。结果:转录组数据结果分析肝纤维化大鼠差异基因富集于丝裂原活化蛋白激酶(MAPK)通路。与对照组相比,模型组ALT、AST含量和MAPK、生长因子β1(TGF-β1)、α平滑肌肌动蛋白(α-SMA)、人I型胶原(COL-I)、白细胞介素6(IL-6)mRNA表达明显升高(P<0.05),白蛋白(Alb)含量明显降低(P<0.05);与模型组相比,冬虫夏草组ALT、AST含量和MAPK、TGF-β、α-SMA、COL-I、IL-6 mRNA表达明显降低(P<0.05),Alb含量明显升高(P<0.05)。结论:冬虫夏草通过调控TGF-β/MAPK通路减轻四氯化碳诱导的大鼠肝脏炎症和纤维化。Objective: To explore the regulatory mechanism of Cordyceps sinensis in improving liver fibrosis.Methods: The differentially expressed genes of liver fibrosis rats and normal rats were analyzed based on the data of RNA SEQ, and the enrichment pathway of differentially expressed genes was found. The rats were randomly divided into control group, model group(CCl4-induced liver fibrosis model) and Cordyceps sinensis group(1 500 mg·kg^(-1)·d^(-1)). After 6 weeks of intragastric treatment, blood from rat’s orbit was used for liver function determination, and RNA from liver was extracted for molecular biology research. Results: Transcriptome analysis showed that the differentially expressed genes were enriched in MAPK pathway. Compared with those in the control group, the contents of ALT and AST, and mRNA expressions of MAPK, TGF-β, α-SMA, COL-I and IL-6 in the model group were significantly increased(P<0.05), and the content of ALB was significantly reduced(P<0.05). Compared with those in the model group, the contents of ALT and AST, and mRNA expressions of MAPK, TGF-β, α-SMA, COL-I and IL-6 in Cordyceps sinensis group were significantly decreased(P<0.05), and the content of ALB was significantly increased(P<0.05). Conclusion: Cordyceps sinensis can regulate the expression of TGF-β/MAPK pathway and attenuate CCL4-induced liver inflammation and fibrosis in rats.
