摘 要:目的:建立超高效液相色谱(UPLC)波长切换法同时测定血栓心脉宁胶囊中6种成分的含量,同时对其进行主成分分析及聚类分析。方法:采用Waters Acquity UPLC HSS T3(50 mm×2.1 mm,1.8μm)色谱柱,流动相为乙腈-0.3%乙酸水溶液梯度洗脱,流速:0.5 ml·min^(-1),检测波长:270,286,296,323,360 nm,柱温:35℃,进样量:3μl;并采用SPSS Statistics 22.0统计软件对含量测定结果进行主成分分析与聚类分析。结果:芦丁、阿魏酸、丹酚酸B、丹参酮ⅡA、华蟾酥毒基和酯蟾毒配基线性范围分别为18.07~451.67μg·ml^(-1)(r=0.9999)、6.07~165.16μg·ml^(-1)(r=0.9998)、8.40~210.04μg·ml^(-1)(r=0.9997)、5.34~133.42μg·ml^(-1)(r=0.9995)、4.77~119.22μg·ml^(-1)(r=0.9986)、3.44~86.03μg·ml^(-1)(r=0.9981);平均加样回收率(n=6)均在98.0%~99.7%,RSD均小于2.0%。主成分分析与聚类分析均可将不同生产厂家的血栓心脉宁胶囊很好的分类,且分类结果一致。结论:本方法快速、准确、重复性好,可用于血栓心脉宁胶囊质量控制。Objective:To establish a UPLC method for the simultaneous determination of 6 constituents in Xueshuan Xinmaining capsules,and to conduct principal component analysis and cluster analysis.Methods:A Waters Acquity UPLC HSS T3(50 mm×2.1 mm,1.8μm)chromatographic column was used;the mobile phase was acetonitrile-0.3%acetic acid solution with gradient elution at the flow rate of 0.5 ml·min^(-1),and the detection wavelengths were 270 nm,286 nm,296 nm,323 nm and 360 nm.The column temperature was 35℃,and the sample size was 3μl.The principal component analysis and cluster analysis were conducted for the results of content determination by SPSS 22.0 software.Results:The linear ranges of rutin,ferulic acid,salvianolic acid B,tanshinoneⅡA,cinobufagin and resibufogenin were 18.07-451.67μg·ml^(-1)(r=0.9999),6.07-165.16μg·ml^(-1)(r=0.9998),8.40-210.04μg·ml^(-1)(r=0.9997),5.34-133.42μg·ml^(-1)(r=0.9995),4.77-119.22μg·ml^(-1)(r=0.9986)and 3.44-86.03μg·ml^(-1)(r=0.9981),respectively.The recoveries ranged from 98.0%to 99.7%and the RSDs were all below 2.0%.The samples could be clearly classified by using CA and PCA,and the results of classification were consistent.Conclusion:The method is fast and accurate with good repeatability,which can be used for the identification and evaluation of Xueshuan Xinmaining capsules,and for the quality control of Xueshuan Xinmaining capsules as well.
