摘 要:目的比较茅苍术生品和麸炒品水提物体外抗心肌细胞缺氧/复氧(H/R)损伤的作用。方法以连二亚硫酸钠诱导H9c2缺氧/复氧损伤模型,MTT法检测细胞活力,ELISA法检测SOD、LDH和MDA的水平;RT-PCR法检测Caspase-3、Bax、Bcl-2 mRNA的表达;Western Blot法检测Caspase-3、Bax、Bcl-2的蛋白表达量。结果与模型组比较,茅苍术生品和麸炒品水提物均能提高细胞的存活率;提高SOD的水平,降低LDH和MDA的水平;提高Bcl-2 mRNA的水平,降低Caspase-3、Bax mRNA的水平;提高Bcl-2的蛋白表达,降低Caspase-3、Bax的蛋白表达量,麸炒品的作用优于生品。结论茅苍术水提物能够体外抗心肌细胞H/R损伤,具有抗氧化和抑制细胞凋亡的作用,麸炒后该作用增强。Objective This study aimed at comparing the protective effects of water extracts from crude and bran-processed Cangzhu(Atractylodis Rhizoma) on H9 c2 cardiomyocyte injured by hypoxia/reoxygenation(H/R). Methods We established an H/R injury model of H9 c2 cardiomyocyte by sodium dithionite(Na2O4S2). The cell viability was detected by the MTT assay. The levels of SOD, LDH and MDA were determined by ELISA, and the mRNA expressions of Caspase-3, Bax and Bcl-2 were determined by quantitative RT-PCR. The expressions of Caspase-3, Bax and Bcl-2 proteins were determined by Western Blot. Results Compared with those of the model group, the cell viability was increased, and the production of SOD was significantly increased. But the productions of LDH and MDA were significantly decreased. The mRNA expression of Bcl-2 was significantly up-regulated, but the mRNA expressions of Caspase-3 and Bax were down-regulated. Meanwhile, the expression of Bcl-2 was stimulated and the expressions of Caspase-3 and Bax were notably inhibited by water extracts obtained from both crude and bran-processed Cangzhu(Atractylodis Rhizoma). The water extract from the processed AL was more effective compared with that from crude Cangzhu(Atractylodis Rhizoma). Conclusion The water extract from Cangzhu(Atractylodis Rhizoma) had the protective effect on H9 c2 cardiomyocyte injured by H/R through anti-oxidation and anti-apoptosis and the effect was enhanced after processed with wheat bran.
