摘  要:目的研究补肾健脾方对去卵巢诱导(OVX)的大鼠骨质疏松的作用及其机制。方法 60只雌性大鼠平均分为6组:假手术组、模型组、中药-低剂量组、中药-中剂量组、中药-高剂量组和西药组。假手术切除大鼠腹腔少量脂肪,其余各组均去除双侧卵巢,其中模型组蒸馏水灌胃,中药组分别低、中、高3个剂量补肾健脾方灌胃,西药组阿仑膦酸钠灌胃。酶联免疫吸附测定(ELISA)检测大鼠血清雌二醇(E2)、I型胶原交联C-末端肽(β-CTX)、Ⅰ型前胶原氨基端原肽水平(PINP),双能X线法检测大鼠腰椎骨密度,压缩实验检测腰椎生物力学,苏木素伊红(HE)染色观察腰椎组织形态学,实时荧光定量PCR(qRT-PCR)检测腰椎破骨调控因子MMP9、CTSK mRNA及成骨调控因子RUNX2、OPG mRNA相对表达。结果模型组大鼠血清E2明显低于假手术组,而β-CTX、PINP明显高于假手术组,中药-高剂量组和西药组β-CTX、PINP均明显低于模型组。模型组大鼠腰椎骨密度、压缩强度均明显低于假手术组,而中药-高剂量组和西药组骨密度、压缩强度均明显高于模型组。模型组MMP9、CTSK表达较假手术组明显上调,中药-高剂量组和西药组MMP9、CTSK表达较模型组均明显下调;模型组RUNX2、OPG表达较假手术组明显下调,中药-高剂量组RUNX2表达较模型组明显上调。结论补肾健脾方可有效防止OVX诱导的大鼠骨量丢失,其机制可能与下调MMP9、CTSK mRNA,上调RUNX2有关。Objective To study the effect and mechanism of Bushen Jianpi Formula(补肾健脾方) on osteoporosis induced by ovariectomy in rats. Methods Sixty female rats were divided into 6 groups on average: sham operation group, model group, low-dose traditional Chinese medicine treatment group, medium-dose traditional Chinese medicine treatment group, high-dose traditional Chinese medicine treatment group and western medicine treatment group. A small amount of fat in the abdominal cavity of the rats was removed by sham operation, and the bilateral ovaries were removed in the remaining groups. The model group was given distilled water by gavage. The traditional Chinese medicine groups were given the corresponding doses of Bushen Jianpi Formula, and the western medicine treatment group was administrated alendronate orally. Serum levels of estradiol(E2), type I collagen cross-linked C-terminal peptide(β-CTX), type I procollagen amino terminal propeptide level(PINP) were detected by ELISA, and dual energy X-ray method was used to detect rat lumbar spine bone mineral density. The compression test was used to examine lumbar vertebra biomechanics. HE staining was used to observe the lumbar spine morphology, and qRT-PCR was used to detect the relative expression of osteoclastogenesis related regulators MMP9, CTSK and osteoblastogenesis related regulator RUNX2, OPG. Results The serum E2 level of the model group was significantly lower than that of the sham operation group, while levels of β-CTX and PINP were significantly higher than those of high-dose traditional Chinese medicine treatment group. The levels of β-CTX and PINP of high-dose traditional Chinese medicine treatment group and the western medicine treatment group were significantly lower than those of the model group.
