摘  要:目的考察南蛇藤提取物对HepG2细胞增殖和黏附作用的影响及可能机制。方法体外培养肝癌细胞HepG2,CCK8法考察南蛇藤提取物对HepG2肝癌细胞增殖的影响;划痕法和Transwell-migration法考察南蛇藤提取物对HepG2肝癌细胞迁移的影响;Transwell-invasion法考察南蛇藤提取物对HepG2肝癌细胞侵袭的影响;黏附实验考察南蛇藤提取物对HepG2肝癌细胞黏附的影响。Western-blot法考察南蛇藤提取物对Wnt/β-Catenin信号通路蛋白的影响。结果 0、15、30、60、120、240μg/mL南蛇藤提取物对HepG2肝癌细胞的48 h抑制率分别为(2.01±1.11)%、(19.68±9.99)%、(28.22±10.13)%、(39.73±7.74)%、(70.91±3.86)%、(77.38±3.01)%,IC50为70.36μg/mL;60μg/mL NST组迁移面积显著低于对照组(t=9.26,P<0.01);60μg/mL NST组迁移细胞数显著低于对照组(t=4.71,P<0.01);60μg/mL NST组侵袭细胞数显著低于对照组(t=2,71,P<0.01);60μg/mL NST组黏附细胞数显著低于对照组(t=7.09,P<0.01);60μg/mL南蛇藤提取物可上调β-catenin、cyclin D1、c-Myc、MMP-9和MMP-2蛋白,下调GSK-3β蛋白。结论南蛇藤提取物可抑制HepG2肝癌细胞的增殖和黏附,其作用机制可能和南蛇藤提取物抑制Wnt/β-Catenin信号通路有关。Objective To investigate the effect of Nansheteng(Celastrus orbiculatus) extract on the proliferation and adhesion of HepG2 cells and its possible mechanism. Methods HepG2 cells were cultured in vitro. CCK8 method was used to investigate the effect of Nansheteng(Celastrus orbiculatus) extract on HepG2 cell proliferation. The scratch method and Transwell migration method were used to investigate the effect of Nansheteng(Celastrus orbiculatus) extract on HepG2 cell migration. Transwell injection method was used to investigate the effect of Nansheteng(Celastrus orbiculatus) extract on HepG2 cell invasion. Adhesion experiment was used to investigate the effect of Nansheteng(Celastrus orbiculatus) extract on HepG2 cell adhesion attached impact. Western blot was used to investigate the effect of Nansheteng(Celastrus orbiculatus) extract on Wnt/β-catenin signaling pathway protein. Results The 48 h inhibition rates of 0 μg/mL, 15 μg/mL, 30 μg/mL, 60 μg/mL, 120 μg/mL and 240 μg/mL extracts of Nansheteng(Celastrus orbiculatus) on HepG2 hepatoma cells were(2.01±1.11)%,(19.68±9.99)%,(28.22±10.13)%,(39.73±7.74)%,(70.91±3.86)% and(77.38±3.01)% and IC50 was 70.36 μg/mL, respectively. The migration area of 60 μg/mL Nansheteng(Celastrus orbiculatus) group was significantly lower than that of the control group(t=9.26, P<0.01). The number of migrating cells in 60 μg/mL Nansheteng(Celastrus orbiculatus) group was significantly lower than that in the control group(t=4.71, P<0.01). The number of invasive cells in 60 μg/mL Nansheteng(Celastrus orbiculatus) group was significantly lower than that in the control group(t=2,71, P<0.01). The number of adherent cells in 60 μg/mL Nansheteng(Celastrus orbiculatus) group was significantly lower than that in the control group(t=7.09, P<0.01). The extract of Nansheteng(Celastrus orbiculatus)(60 μg/mL) can up regulate the protein of β-Catenin, cyclin D1, c-myc, MMP-9 and MMP-2, and down regulate the protein of GSK-3β.
