摘  要:目的筛选治疗急性胰腺炎的大黄活性成分、活性成分靶基因,探讨靶基因的生物学功能及相关信号通路。方法依托TCMSP数据库查询大黄所对应的靶基因,通过ADME参数(DL≥0.18,OB≥30%)筛选大黄可能的活性成分及其靶基因;然后,运用Genecards、OMIM、DRUGBANK等数据库获取急性胰腺炎的发病相关基因;并利用R语言将大黄活性成分靶基因与急性胰腺炎发病相关特异性基因交集,进而得到关键靶基因;通过Metascape平台进行关键靶基因生物学功能分析(GO注释和KEGG富集)。结果 TCMSP数据库中检索到大黄活性成分共92个,通过ADME参数筛选到的活性成分主要包括β-谷甾醇、芦荟大黄素等,活性成分靶基因主要有NCOA2、PTGS2、PIK3CG、BCL2、BAX等。关键靶基因共包括CASP3、BAX、BCL2等30个,主要参与药物反应、凋亡信号通路、细胞增殖负调控等生物过程,膜筏、膜微域、膜域等重要的细胞组分,G蛋白偶联乙酰胆碱受体活性、G蛋白偶联胺受体活性、乙酰胆碱受体活性等分子功能;其富集在癌症、细胞凋亡、IL-17等多条信号通路上。结论治疗急性胰腺炎的大黄活性成分主要包括β-谷甾醇、芦荟大黄素等,活性成分靶基因主要有NCOA2、PTGS2、PIK3CG、BCL2、BAX等;关键靶基因主要参与药物反应、凋亡信号通路、细胞增殖负调控等生物学过程,相关通路主要有癌症、细胞凋亡、IL-17等信号通路。Objective acute pancreatitis,and to explore the biological functions and related pathways of the target genes of the rhubarb.Methods screened out by ADME parameters(DL≥0. 18,OB≥30%). Then,using the Genecards database,OMIM database,DRUGBANK databases to obtain the disease-related targets of acute pancreatitis. Using the R language to link the target genes of the rhubarb to the acute pancreatitis,and then we obtained the key target genes. Finally,we performed the biological function analysis of potential common target genes(GO annotation and KEGG enrichment)through the Metascape platform.Results ents screened by ADME parameters mainly included β-sitosterol and aloe-emodin,etc. The target genes of active ingredients mainly included NCOA2,PTGS2,PIK3 CG,BCL2,BAX,and other genes. There were 30 key target genes,mainly including CASP3,BAX,BCL2,etc.,which were mainly involved in the biological processes such as drug reaction,apoptosis signaling pathway,negative regulation of cell proliferation,and cellular components such as membrane raft,membrane microdomain,membrane region,and molecular functions such as G-protein-coupled acetylcholine receptor activity,G-protein-coupled amine receptor activity,and acetylcholine receptor activity. These key target genes were enriched in multiple signaling pathways such as pathways in cancer,apoptosis,IL-17,etc.Conclusions of rhubarb in the treatment of acute pancreatitis include β-sitosterol,aloe-emodin,etc. The target genes of active ingredients mainly include NCOA2,PTGS2,PIK3 CG,BCL2,BAX,etc. These key target genes are involved in the biological processes such as drug reaction,apoptosis signaling pathway,negative regulation of cell proliferation,etc.;the related pathways mainly include pathways in cancer,apoptosis,IL-17,etc.
