摘  要:为研究荞麦蜜对酒精引起的肝损伤和肠道菌群紊乱的改善效果,对酒精诱导形成的肝损伤小鼠饲喂荞麦蜜予以干预后,测定肝脏指数、血清转氨酶活力以及分析对肝脏脂代谢、氧化应激、肝脏组织的影响,并结合高通量测序分析小鼠的肠道菌群变化。结果显示,与酒精模型组相比,荞麦蜜能显著降低酒精性肝损伤小鼠的谷丙转氨酶(alanine aminotransferase,ALT)、谷草转氨酶(aspartate aminotransferase,AST)、丙二醛(malondialdehyde,MDA)水平(P<0.05),缓解肝脏甘油三酯(triglyceride,TG)含量升高,提高超氧化物歧化酶(superoxide dismutase,SOD)活力,明显减少了酒精引起的肝脏脂滴现象,脂肪变性情况得到明显改善。而且,相比于酒精模型组,荞麦蜜的摄入改善了酒精摄入引起的肠道菌群多样性和丰富度的变化,拟杆菌门相对丰度升高7.18%,厚壁菌门相对丰度降低15.59%,厚壁菌门与拟杆菌门的比值降低21.25%,恢复到空白对照组水平;进一步在属水平上分析了相对丰度排前36的微生物,发现荞麦蜜摄入明显改善了酒精引起的16种菌属的变化。结果表明荞麦蜜能改善酒精引起的小鼠肝损伤以及肠道菌群的失衡,可以作为一种保肝护肝产品的原料加以开发和利用。In order to study the effect of buckwheat honey on improving the liver injury induced by alcohol and the dysbiosis of gut microbiota in mice,the liver index,activity of serum transaminase was measured after buckwheat honey administration in the mice with liver injury induced by alcohol.The effects of the liver index,activity of serum transaminase on the liver lipid metabolism,oxidative stress and liver tissue were analyzed.The changes of gut microbiota in mice were analyzed with high-throughput sequencing.The results showed that compared with the alcohol model group,buckwheat honey significantly reduced the levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),malondialdehyde(MDA),and the liver fat droplet phenomenon caused by alcohol(P<0.05),reduced the level of liver triglyceride(TG),improved the activity of superoxide dismutase(SOD),and significantly improved the steatosis.Compared with the alcohol model group,the intake of buckwheat honey improved the diversity and richness of gut microbiota in mice caused by alcohol intake.At the phylum level,the abundance of Bacteroidetes increased by 7.18%,the abundance of Firmicutes decreased by 15.59%and the ratio of Firmicutes to Bacteroidetes decreased by 21.25%and restored to the level of blank control group.The results of further analyzing the top 36 abundant microorganisms at the genus level showed that buckwheat honey significantly improved the changes of 16 bacterial genera caused by alcohol.It is indicated that buckwheat honey can improve the liver injury and gut microbiota imbalance caused by alcohol in mice,and can be developed and utilized as a raw material for liver protection products.
