摘 要:目的探讨转化生长因子-β1(TGF-β1)基因单核苷酸多态性(SNP)与系统性红斑狼疮肺间质病变(SLE-ILD)患者肺高分辨率计算机断层扫描(HRCT)影像学表现的关联。方法搜集符合纳入标准的SLE-ILD患者共80例,按照HRCT影像学表现分为斑片状、磨玻璃影、网格状、条索状4类。采用Sequenom基因分型实验技术对TGF-β1基因的两个SNPs位点rs12985162、rs10417924分型,并对基因型和等位基因与肺HRCT影像学表现进行相关性分析。结果在rs12985162,C等位基因在磨玻璃影组中显著高于非磨玻璃影组,差异有统计学意义(P=0.01);T等位基因和TT基因型在磨玻璃影组显著低于非磨玻璃影组,差异具有统计学意义(P=0.01,P=0.021)。在位点rs10417924,T等位基因频率在磨玻璃影组显著高于非磨玻璃影组,C等位基因在磨玻璃影组显著低于非磨玻璃影组,差异有统计学意义(P=0.047,P=0.047);在位点rs12985162、rs10417924,TGF-β1 SNPs的等位基因和基因型在斑片状与非斑片状,条索状与非条索状,网格状与非网格状进行比较,均未见统计学差异。结论TGF-β1两个位点rs12985162、rs10417924可能与SLE-ILD患者肺HRCT磨玻璃影表现有关。Objective To investigate the association between gene single nucleotide polymorphism(SNP)of transforming growth factor-β1(TGF-β1)and high resolution CT(HRCT)imaging in patients with systemic lupus erythematosus interstitial pulmonary disease(SLE-ILD).Methods A total of 80 patients with SLE-ILD who met the inclusion criteria were divided into four types:patchy,ground glass shadow,grid and cord according to the imaging findings of HRCT.The rs12985162 and rs10417924 genotypes of two SNPs sites of TGF-β1 gene were analyzed by Sequenom genotyping technique,and the correlation between genotypes,alleles and pulmonary HRCT imaging findings was analyzed.Results In rs12985162,the C allele in the ground glass group was significantly higher than that in the non-ground glass group,and there was a statistical difference(P=0.01);the T allele and TT genotype in the ground glass group were significantly lower than those in the non-ground glass group and there was a statistical difference(P=0.01,P=0.021).At locus rs10417924,the frequency of the T allele in the ground glass group was significantly higher than that in the non-ground glass group,and the C allele in the ground glass group was significantly lower than that in the non-ground glass group,with statistical differences(P=0.047,P=0.047);At sites rs12985162 and rs10417924,the alleles and genotypes of TGF-β1 SNPs are compared in patchy and non-patchy,cord-shaped and non-striped cord-shaped,and grid-shaped and non-grid-shaped.There was no statistical difference.Conclusion The two sites of TGF-β1,rs12985162 and rs10417924,may be related to the HRCT ground-glass opacity in patients with SLE-ILD.
