摘  要:目的研究人类精子囊性纤维化跨膜传导调节因子(CFTR)表达部位、分子量以及不同活力精子的表达水平。方法免疫组化法检测正常人类精子CFTR表达部位,蛋白印迹法检测正常人精子和大鼠睾丸组织CFTR蛋白,研究其分子量水平;30份人精液按精子前向运动率(PR)梯度分组,分析不同活力精子的CFTR表达水平。结果人类精子CFTR蛋白分子量以75kd为主,主要表达于精子尾部的中段,与精子线粒体的分布部位是一致的;精子的活力与75kd CFTR蛋白表达量呈阶梯式正相关(P<0.01)。结论人类精子CFTR蛋白主要表达于精子尾部,其活动可能与精子的能量代谢有关,并参与了精子活力的调节机制。Objective To investigate the expression level of CFTR and its distribution in human different motile sperms. Methods The expression distribution of CFTR in normal human sperm was determined by immunohistochemistry,and the protein expression levels of CFTR in normal human sperm and rat testicular tissue were detected by western blot, and the changes in CFTR expressions were comparatively analyzed among the groups with different PR sperm gradient groups.Results The molecular weight of CFTR protein in human sperm was about 75 kD.CFTR protein was distributed at the middle segment of sperm tail, which corresponds to the location of sperm mitochondria. There was a step-wise positive correlation between sperm motility and CFTR protein expression. Conclusion Human sperm CFTR protein is mainly expressed in the tail of sperm, indicating that the activity of CFTR protein may be related to sperm energy metabolism and involved in the regulation mechanism of sperm motility.
