摘  要:目的:考察川楝子对大鼠内源性代谢物的影响,初步探讨其亚急性毒性代谢途径和可能机制。方法:以川楝子为示例药物,以大鼠为研究对象,随机分为川楝子高剂量组、川楝子中剂量组、川楝子低剂量组并给予相应剂量药液灌胃,正常组给予等量生理盐水,连续给药21 d。通过代谢组学技术对采集到的大鼠血清、肝、肾样本进行检测,用主成分分析(PCA)结合正交偏最小二乘判别分析(OPLS-DA)进行代谢差异表征,寻找差异代谢物及可能的代谢通路。结果:代谢组学分析显示,组间代谢差异明显,与正常组比较,川楝子组血清、肝、肾样本中分别鉴定出10、20和12个差异代谢物,这些代谢物水平存在明显的变化。结论:川楝子具有一定的肝毒性和潜在肾毒性,其机制可能与ABC转运蛋白、氨基酸代谢、嘌呤和嘧啶代谢等有关。Objective:To observe the effect of Fructus Toosendan on endogenous metabolites of rats,thus to explore the metabolic pathway and possible mechanism of its sub-acute toxicity.Methods:32 SD rats were randomly divided into the normal group,the Fructus Toosendan groups of high-dose,medium-dose and low-dose,with 8 rats in each group.After 21 days of continuous corresponding administration,the samples of serum,liver and kidney were collected for detection.Principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)were used to identify the potential biomarkers and possible metabolic pathways.Results:The results of metabolomics analysis showed that there were 10,20 and 12 different metabolites in the samples of serum,liver and kidney respectively compared to those in the normal group,and there were significant changes in the levels of these metabolites.Conclusion:Fructus Toosendan has certain liver toxicity and renal toxicity,and its mechanism may be related to ABC transporters,metabolisms of amino acids,purine and pyrimidine.
