摘  要:为解析花生产量遗传基础,挖掘稳定存在的控制花生籽仁大小相关性状的QTL(quantitative trait locus),本研究以花育28号和P76作为亲本,构建包含146个重组自交家系(recombination inbred line,RIL)的RIL作图群体,进行籽仁大小相关性状的QTL定位分析。利用SSR和特定位点扩增长度测序(SLAF-seq)技术,构建了一张包含2350个SLAF标签、61个SSR标记、20个连锁群、覆盖长度为1814.72 cM、平均间距为0.76 cM的遗传连锁图谱。结合本研究构建的遗传图谱,利用QTL IciMapping v4.1对两个环境下籽仁大小相关性状进行QTL定位分析,共检测到8个控制籽仁长宽比的QTL、7个控制籽仁长的QTL、5个控制籽仁宽的QTL,QTL的加性效应对表型的总贡献率分别为63.153%、42.227%和32.496%。A02染色体上Marker10593~Marker97229区间同时定位到控制籽仁长(qSLA02.1)、籽仁宽(qSWA02)和籽仁长宽比(qSLWA02.1)相关QTL、Marker140970~Marker133566区间同时定位到控制籽仁长(qSLA02.2)和籽仁长宽比(qSLWA02.2)相关QTL。本研究结果为挖掘花生籽仁大小性状基因和分子育种奠定了基础。To analyze the genetic basis for seed size of peanut and explore the stably existed QTLs that controlling the peanut seed size,a mapping population of 146 lines(recombination inbred lines,RIL),derived from a cross between Huayu28 and P76,was applied to analyze QTLs about seed size.The specific locus amplified fragment sequencing(SLAF-seq)technology was employed to construct a high-density genetic map,which contained 2,350 SLAF,61 SSR,20 linkages,covered 1,814.72 cM and had an average distance of 0.76 cM between adjacent markers.Based on the genetic map,QTL mapping analysis for seed size related traits under two environments was performed with QTL IciMapping v4.1.Multi-environ-mental joint analysis demonstrated that the collective PVEs by additive effect for seed length(SL),seed width(SW)and ratio of seed length to width(SLW)were 63.153%,42.227%and 32.496%,respectively.The Marker10593-Marker97229 interval on A02 harbored three QTLs,i.e.qSLA02.1 for SL and qSWA02 for SW and qSLWA02.1 for SLW,the Marker140970-Marker133566 interval on A02 harbored two QTLs,i.e.qSLA02.2 for SL and qSLWA02.2 for SLW.These results laid the foundation for further explo-ring candidate genes toward seed size and molecular breeding.
