摘  要:[目的]研究蝉拟青霉产镇痛组分的最佳液体发酵培养条件。[方法]采用Plackett-Burman试验设计对液体培养基中葡萄糖、蛋白胨、酵母浸膏、KH2PO4和MgSO4以及发酵过程中p H、温度和接种量8个因素进行变量分析,筛选出了重要的影响因子为培养基p H、发酵温度和接种量;通过最陡爬坡试验得到这些因子的最大响应值区域。[结果]应用响应面方法分析得到最佳培养条件为:葡萄糖20 g/L、蛋白胨3 g/L、酵母浸膏2 g/L、KH2PO41 g/L、MgSO40.5 g/L、p H 7.96、温度16.05℃及接种量9.27%。[结论]优化后得到的镇痛组分含量是原条件的7.633倍,为该菌株镇痛组分的工业化发酵生产奠定了较好的基础。[Objective]To study the optimal culture conditions for extracting analgesic components from the liquid fermentation of Paecilomyces cicadae.[Method]the eight-variable analysis of glucose,peptone,yeast extract,KH2 PO4,MgSO4,p H,temperature,and inoculum were analyzed with the Plackett-Burman test design,and the important factors,such as p H,temperature,and inoculum were selected out from 8 factors.Then the area of the maximum response value of factors were obtained through the steepest climbing test,and the maximum response value of factors were analyzed by the response surfaces method.[Result]The optimal culture conditions were glucose 20 g/L,peptone 3 g/L,yeast extract 2 g/L,KH2 PO41 g/L,MgSO40.5 g/L,p H 7.96,temperature 16.05℃and inoculation volume 9.27%.[Conclusion]The content of analgesic components obtained after optimization is 7.633 times of the original conditions.After optimization,it adds a better foundation for the industrialized fermentation production of the baseline analgesic component.
