摘  要:对甜荞根转录组的SSR位点进行了统计分析,并设计了引物。利用Misa软件在长度大于200 bp的55392条Transcripts中,共检索到3906个SSR位点,出现频率为1/1.709 kb。随着Transcripts长度的增加,含SSR序列的比例和SSR分布频率均增加。单、二和三核苷酸重复类型共占SSR位点总数的95.80%,其中单核苷酸重复数量最多,占48.13%;(A)_(n)、(AG)_(n)和(AAG)_(n)分别是单、二和三核苷酸重复的优势基序,分别占47.51%、7.65%和9.49%。SSRs数量随着基序重复次数的增加而减少。10次(25.45%)、6次(15.34%)和5次(20.71%)重复的SSR数量最多。基于Krait v0.10.2软件共成功设计了1183对SSR引物,其中3对来自黄酮生物合成关键基因。SSR loci were analyzed and primers were designed based on root transcriptome of common buckwheat.A total of 3906 SSRs were detected in 55392 transcripts(length>200 bp)by Misa software,with the mean distance of 1.709 kb between SSRs.The mean distance between SSRs and portion of transcripts with SSRs increased with the length of transcript increased.Most of the SSR types were mononucleotide,dinucleotide and trinucleotide repeat,accounting for 95.80%of total SSRs.(A)_(n),(AG)_(n)and(AAG)_(n)were most abundant motifs for mononucleotide,dinucleotide and trinucleotide,accounting for 47.51%,7.65%and 9.49%of total SSRs,respectively.The number of SSRs decreased with the repeat number increased,and 10,6 and 5 tandem repeats were most abundant repeats,accounting for 24.24%,15.34%and 20.71%of total SSRs,respectively.Totally,1183 pairs of SSR primers were successfully designed by Krait v0.10.2 software,among which 3 pairs were derived from the key genes involved in flavonoid biosynthesis.
