摘  要:为明确太岁中真菌菌群结构及分离真菌菌株存在的真实性,在太岁组织不同层次重复取样,采用热溶解膜过滤法处理后提取DNA,Illumina Miseq高通量测序,并进一步将从太岁中分离的腐皮镰刀菌(Fusarium solani)与测序结果中的同属OTU做相似性比对.结果表明:供试太岁所含真菌多样性丰富,包括12个门56个目146个属;真菌分布不均匀,内层多样性高于外层;相对丰度≥1%的内外层共有属分别是以土壤为生境的虫草属(Cordyceps)、曲霉科未分类属(unclassified Aspergillaceae)、踝节菌属(Talaromyces),占比之和为11.13%~25.19%;分离的腐皮镰刀菌在测序数据中能比对到相似性为100%的OTU802.可见,太岁中真菌不存在共同优势菌群,对太岁形成作用有限,但能印证其生境为土层;太岁中腐皮镰刀菌及一些常见真菌真实存在.To identify the fungi species and understand the community structure in Taisui,tissues were sampled at different layers of Taisui.The DNA was extracted after thermolysis membrane filtration,and sequenced by Illumina Miseq high-throughput sequencing.Then similarity between Fusarium solani isolated from Taisui and the OTU of the same genus from the sequencing results were compared.The results showed that the fungi detected in Taisui had a rich diversity,including 12 phyla,56 orders and 146 genera.The distribution of the fungi was uneven,which was more diverse in the inner layer and less in the outer layer.The common genera with relative abundance higher than 1% were all soil borne fungi,including Cordyceps,unclassified Aspergillaceae and Talaromyces,with relative abundance totaling between 11.13% and 25.19%.The ITS sequence of F.solani isolated from Taisui shared 100%similarity with that of OTU802 in sequence alignment.It can be concluded that there was no common dominant fungi community in Taisui,which exerted very limited effect on its development;but it can confirm that the soil was the habitat of Taisui;and F.solani and other fungi commonly existed in Taisui.
