摘 要:目的比较不同肌性组织在三种冰冻条件下的切片效果,筛选出不同肌性组织的最优冰冻方法。方法采用骨骼肌(腓肠肌)、心肌、平滑肌(胸主动脉);三种冰冻条件分别为液氮浸入法、液氮漂浮法、直接-80℃法。通过取材,冰冻,切片和苏木精-伊红(hematoxylin-eosin,HE)染色,最后在镜下观察组织结构形态。结果骨骼肌冰冻的最优条件是液氮浸入法,心肌冰冻的最优条件是液氮漂浮法,血管平滑肌冰冻的最优条件是液氮浸入法或液氮漂浮法。结论不同肌性组织最适宜的冻存条件存有差异。Objective To select the optimal freezing method for different muscular tissues by comparing the effect of different muscular tissue sections under three freezing conditions.Methods The tissues used in this experiment included skeletal muscle(gastrocnemius),myocardium,and vascular smooth muscle(thoracic aorta).The three freezing conditions were liquid nitrogen immersion,floating on liquid nitrogen,and direct-80℃methods.After sampling,freezing,sectioning and hematoxylin-eosin(HE)staining,the tissue structure morphology was observed under a microscope.Results The optimal freezing condition for the skeletal muscle,myocardium and vascular smooth muscle was liquid nitrogen immersion,floating on liquid nitrogen,and liquid nitrogen immersion or floating on liquid nitrogen,respectively.Conclusion There are differences in the optimal freezing conditions for different muscular tissues.
