摘  要:目的:探讨中性粒细胞胞外诱补网(NETs)抗痤疮丙酸杆菌的作用机制。方法:提取正常人外周血多形核中性粒细胞(PMNs),并使其与痤疮丙酸杆菌进行孵育。采用荧光共聚焦倒置显微镜(confocal)观察NETs的形成,使用SYTOX Green对NETs的形成进行定量,菌落计数法检测NETs对痤疮丙酸杆菌的杀伤。结果:痤疮丙酸杆菌可以诱导NETs的形成,随着时间的延长,NETs的形成逐渐增多(90 min,15%;120 min,22%;180 min,44%;P<0.05);在PMNs与菌孵育时间固定的基础上(2 h),随着痤疮丙酸杆菌菌量的增多(1∶5,1∶10,1∶20),NETs的产生明显增多(11%、21%、43%;P<0.05);中性粒细胞对痤疮丙酸杆菌具有杀伤作用,中性粒细胞对痤疮丙酸杆菌的胞外杀伤机制主要通过NETs介导。结论:中性粒细胞胞外诱捕网NETs在杀伤痤疮丙酸杆菌过程中发挥着关键作用。Objective: To investigate the role of neutrophil extracellular traps(NETs) against Propionibacterium acnes(P. acnes). Methods: Polymorphonuclear neutrophils(PMNs) were isolated from healthy blood donors and subsequently infected with P. acnes. By assessing the co-localization of DNA and neutrophil elastase, we confirmed NETs formation entrapping P. acnes specimens. And then SYTOX Green was used to stain these NETs after PMNs infected with P. acnes for quantifying NETs formation. By colony-forming units assay, the ability of NETs to kill P. acnes was detected. Results:Our results demonstrated that P. acnes could trigger NETs formation, and the production of NETs was triggered in a time-dependent manner upon exposure to P. acnes, in both time-dependence(90 min, 15%;120 min, 22%;180 min, 44%;P<0. 05) and in dose dependence(1∶5, 11%;1∶10,21%;1∶20, 43%;P<0. 05). Moreover, NETs had killing effect on P. acnes. Conclusion: Our results indicate that NETs may have crucial innate immune effect against P. acnes infection.
