摘 要:目的探讨亚抑菌浓度β-内酰胺类抗菌药物对金黄色葡萄球菌临床株脂蛋白样蛋白(Lpl)的诱导作用。方法遴选我国主要流行克隆群CC239(ST239)中的菌株作为实验对象,在检测菌株抗生素敏感性的基础上,采用亚抑菌浓度苯唑西林(OXA)进行处理,十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)分析菌体蛋白;制备重组Lpl-His蛋白及其多克隆血清,采用Western blot对诱导蛋白进行鉴定。结果遴选的6株ST239型临床分离的耐甲氧西林金黄色葡萄球菌(MRSA),抗菌药物敏感性检测显示t030和t037菌株对利福平和复方新诺明的敏感性呈相反趋势,均可在亚抑菌浓度OXA诱导下产生约30 kDa的蛋白质,经制备脂蛋白Lpl抗体,Western blot鉴定该受OXA诱导表达的蛋白质为金黄色葡萄球菌Lpl,其诱导作用具有剂量依赖效应。结论亚抑菌浓度苯唑西林可诱导金黄色葡萄球菌MRSA临床分离株产生Lpl,该诱导蛋白在金黄色葡萄球菌致病性中的作用值得深入研究。Objective To evaluate the induction role of subinhibitory concentrations ofβ-lactams in the production of Staphylococcus aureus lipoprotein-like protein(Lpl).Methods We selected the most predominant multidrug-resistant clonal strains of sequence type(ST)239 with SCCmec typeⅢmethicillin-resistant S.aureus(ST239-MRSA-Ⅲ)prevailed in China as subjects and then tested their susceptibility against antibiotics.After that,all the operations involved were carried out on the basis of detecting the antibiotic sensitivity of the strain.SDS-PAGE were used to analyze the bacterial proteins of selected strains post-treated with subinhibitory concentrations of oxacillin.Western blot were used to identify the prepared recombinant Lpl-His proteins and the polyclonal antibodies against Lpl-His.Results Six strains of ST239 methicillin-resistant Staphylococcus aureus(MRSA)isolated from clinic were selected in the experiment.Compared with t030 ST239-MRSA-Ⅲstrains,t037 strains showed opposite resistance to rifampicin and trimethoprim/sulfamethoxazole in the antimicrobial susceptibility test.However,both types of ST239-MRSA can produce a 30kDa protein under the induction of the subinhibitory concentrations of oxacillin.The protein induced by oxacillin was demonstrated as Staphylococcus aureus Lpl by Western blot,and its induction had dose-dependent effect.Conclusion The subinhibitory concentrations of oxacillin can induce the production of S.aureus Lpl,whereas the role of this induced protein in the pathogenicity of S.aureus infection in vivo is worth of future investigation.
