摘  要:翻译控制肿瘤蛋白(translational controlled tumor protein.TCTP)广泛分布在真核生物中,其表达在翻译和转录水平上受控制。近年来,在植物中对TCTP研究集中在植物生长与抗逆性方面。该试验以PVY高感品种“NC95”和高抗品种“SY04-3”为材料,克隆TCTP基因,并进行生物信息学以及相对表达量分析。结果表明:从“SY04-3”和“NC95”中分离出来的TCTP基因均包含507 bp的ORF,二者基因序列存在7处碱基突变,但均编码168个氨基酸,氨基酸序列存在1个区别。通过生物信息学分析显示,“SY04-3”和“NC95”分子量分别是18782.16和18773.06 kD,等电点分别是4.33和4.25。通过对氨基酸分析发现,TCTP蛋白为非跨膜亲水性蛋白,无信号肽,可能定位在细胞质,属于TCTP家族蛋白,“SY04-3”和“NC95”分别有13个和14个磷酸化位点,二者均有1个糖基化位点,进化分析显示与辣椒和茄子的亲缘关系最近。Translationally controlled tumor protein(TCTP)is widely distributed in eukaryotes and its expression is controlled at the translation and transcription levels.In recent years,studies on TCTP of plants have focused on plant growth and stress resistance.In this study,the TCTP genes were cloned from PVY sensitive variety“NC95”and PVY resistant variety“SY04-3”.Bioinformatics and relative expression of TCTP were further analyzed.The results showed that the TCTP genes from both“SY04-3”and“NC95”contained 507 bp of ORF with seven base mutations,but encoded 168 amino acids,with one difference in sequence.Bioinformatic analysis showed that the molecular weight and isoelectric point of TCTP from“SY04-3”were 18782.16 kD and 4.33,respectively,and those of TCTP from“NC95”were 18773.06 kD and 4.25,respectively.Amino acid analysis showed that TCTP protein was a non-transmembrane hydrophilic protein without signal peptide,which might be located in the cytoplasm and belonged to TCTP familyproteins.Furthermore,TCTP from“SY04-3”and“NC95”had 13 and 14 phosphorylation sites,respectively,and both TCTP had one glycosylation site.Phylogenetic analysis showed that TCTP protein was most closely related to pepper and eggplant.
