摘　　要：目的建立高效液相色谱法同时测定红旱莲中芦丁、金丝桃苷、槲皮素、异槲皮苷、山柰苷的含量并研究其变化。方法采用高效液相色谱法在360 nm下,对28批不同产地的红旱莲进行测定,并对5批河南南阳产红旱莲的根、茎、叶、花、果实、地上部分中5个黄酮类成分进行含量测定。结果芦丁、金丝桃苷、槲皮素、异槲皮苷、山柰苷质量浓度分别在26.9~403.5、21.37~320.55、32.72~490.8、26.73~400.95、17.7~265.5μg·mL-1与峰面积呈良好的线性关系(r均>0.9998);5个黄酮类成分的平均加样回收率均在98.27%~103.72%,RSD在0.43%~0.75%。结论不同产地红旱莲中黄酮类成分含量差异较大,其中槲皮素含量最高;8月份左右果成熟后采收总黄酮含量最高;不同部位总黄酮含量比较如下:果>茎>地上部分>花>叶>根。该方法简便、准确,结果稳定,可用于红旱莲中黄酮类成分的含量测定和质量评价。Objective To simultaneously determine rutin,hyperoside,quercetin,isoquercetin and kaempferol in Hypericum ascyron L.by HPLC,and study the content changes.Methods The content of 28 batches of Hypericum ascyron L.from different areas and the content of 5 flavonoids in the roots,stems,leaves,flowers,fruits and parts above the ground of 5 batches of Hypericum ascyron L.from Nanyang,Henan were determined at 360 nm by HPLC.Results The detection concentrations of rutin,hyperin,quercetin,isoquercetin and kaempferol were at 26.9~403.5,21.37~320.55,32.72~490.8,26.73~400.95 and 17.7~265.5μg·mL-1,respectively,with a good linearity in the peak area(all r>0.9998).The average recovery was 98.27%~103.72%,and RSD was 0.43%~0.75%.Conclusion The content of Hypericum ascyron L.in different areas is quite different,but the content of quercetin is the highest.The total content of fruits harvested in August is the highest.The total content in different parts follows:fruits>stems>parts above the ground>flowers>leaves>roots.The method is simple,accurate and stable,which can be used for the content determination and quality evaluation of flavonoids in Hypericum ascyron L.
