摘  要:为提高普通小麦染色体微切割制片质量,以中国春小麦为实验材料,进行染色体制片、单个二价体染色体微切割和微收集研究。结果表明,先将载玻片放在垫片装置上,使片膜和载玻片置于垫片装置的上玻片上,MMI载玻片置于垫片装置的下玻片上,再轻压盖片,花粉母细胞和二价体染色体就容易分散,分散效果较好,利于二价体染色体微切割、微收集,较好地解决了花粉母细胞及二价体染色体聚在一起不容易分散的难题,为植物染色体微切割微收集技术广泛应用提供技术支持。In order to improve the making sheet quality of micro-cutting of common wheat chromosomes,using ordinary Chinese Spring wheat as material,research on chromosome making sheet,bivalent micro-cutting and micro-collection.The results show that when placing the MMI glass sheet on the gasket device that we designed such that the slide with the film is placed on the upper slide of the gasket device and MMI glass sheet is placed on the lower glass sheet of the gasket device,then gently pressing the cover glass sheet,the pollen mother cells and the divalent chromosomes are more easily dispersed and the dispersion effect is better.This technic solves the dispersion problem and facilitates the Micro-cutting and micro collection of the divalent chromosomes and better solves the dispersion problem.
