摘　　要：为研究不同乳脂含量水平的德宏奶水牛乳腺组织中H-FABP基因的表达差异,本试验采集216头德宏奶水牛的乳样,用乳成分分析仪测定乳脂率,将其中乳脂率在7%~8%的定为中乳脂率组(M组),高于8%的为高乳脂率组(H组),低于7%的为低乳脂率组(L组),并选取同一养殖小区、饲养条件和胎次相同、年龄相仿、泌乳盛期的不同乳脂率的德宏奶水牛各3头,采集乳腺组织,利用实时荧光定量PCR技术检测H-FABP基因的mRNA表达水平,并与乳脂率进行相关性分析。结果显示,H-FABP基因mRNA表达水平H组极显著高于M组和L组(P<0.01),M组极显著高于L组(P<0.01)。相关性分析表明,H-FABP基因表达量与乳脂率呈极显著正相关(P<0.01)。结果表明,H-FABP基因的表达影响乳脂的合成。In order to study the expression differences of heart fatty acid-binding protein(H-FABP)gene in the mammary tissues of Dehong dairy buffalo with different milk fat contents,216 Dehong dairy buffalo were collected in this experiment and milk samples were selected for detection the milk fat content with milk component analyzer.The milk fat content of most Dehong dairy buffalo was 7.5%±0.5%,the milk fat content was between 7%to 8%in the middle milk fat content group(M group),and higher than 8%was the high milk fat content group(H group).Less than 7%was the low milk fat content group(L group).There were 3 healthy Dehong milk buffaloes in each groups,which were the same feeding area,the similar breeding conditions and parity,the same age,all animals were selected at same period of lactation,milk samples were collected to detected the milk fat content.H-FABP gene mRNA expression level in the mammary tissues tested by real-time PCR.The correlation between the H-FABP gene expression and milk fat content were analyzed.The results showed that the mRNA expression level of H-FABP gene in group H was extremely significantly higher than that of group M and L(P<0.01),group M was extremely significantly higher than group L(P<0.01).Correlation analysis showed that the expression of H-FABP gene was significantly positively correlated with milk fat content(P<0.01).The results indicated that the expression of H-FABP gene affects milk fat content.
