摘  要:目的:探讨在高碘状态下胎盘细胞中绒毛膜促性腺激素(chorionic gonadotropin,HCG)对钠碘转运体(sodium iodide symporter,NIS)和pendrin的调节作用。方法:将人早孕胎盘绒毛滋养细胞株(HPT-8)于DMEM培养基中培养。分别设置高碘组(5000μg/L、500μg/L)和对照组,高碘(5000μg/L、500μg/L)+HCG(0.5 U/ml、5 U/ml)组。对照组和高碘组细胞培养48 h,高碘+HCG组在高碘培养24 h后加入HCG继续培养24 h,各组分别提取RNA,使用实时荧光定量PCR检测NIS mRNA和pendrin mRNA的表达量。结果:5000μg/L高碘组和500μg/L高碘组的NIS mRNA和pendrin mRNA表达量高于对照组(P<0.01);5000μg/L高碘状态下,0.5 U/ml与5 U/ml HCG组的NIS mRNA和pendrin mRNA表达量均显著低于未加入HCG组(P<0.05);高碘和HCG对NIS mRNA和pendrin mRNA的表达不产生交互作用(R^(2)=0.836,F=0.087,P=0.776;R^(2)=0.325,F=2.970,P=0.123)。结论:高碘状态下HCG可降低胎盘细胞的摄碘能力。Objective:To investigate the regulation of chorionic gonadotropin(HCG)on sodium iodide symporter(NIS)and pendrin in human trophoblast cells(HPT-8)under high iodine status.Methods:After cultured with DMEM,HTP-8 cells were divided into blank group,pure high iodine group(5000μg/L,500μg/L)and high iodine(5000μg/L,500μg/L)+HCG(0.5 U/ml,5 U/ml)group,respectively.The blank and pure high iodine group were cultured for 48 h,the high iodine+HCG group was cultured in high iodine state for 24 h and the HCG was added to continue the culture for 24 h.RNA was extracted,and real-time quantitative PCR was performed to detect the expression levels of NIS mRNA and pendrin mRNA.Results:The expressions of NIS mRNA and pendrin mRNA in the 5000μg/L high iodine group and the 500μg/L high iodine group was respectively higher than those of the blank group(P<0.05).Under the condition of 5000μg/L high iodine,the expression levels of NIS mRNA and pendrin mRNA in both 0.5 U/ml and 5 U/ml HCG groups were lower than those in the control group(P<0.05).Different levels of iodine and HCG have no interaction effect on the expressions of NIS mRNA and pendrin mRNA(R^(2)=0.836,F=0.087,P=0.776 and R^(2)=0.325,F=2.970,P=0.123).Conclusion:With the participation of HCG,the expressions of NIS mRNA and pendrin mRNA in HPT-8 cells decrease,indicating that HCG can reduce the iodine uptake capacity of HPT-8 cells under high iodine state.
