摘 要:细胞治疗作为肿瘤学研究和临床实践中的新型疗法,通过修复受损组织或替换身体缺陷细胞,在多种疾病治疗中发挥着越来越重要的作用。但对细胞治疗产品治疗疾病的机制和细胞移植后的宿主体内的分布和迁移情况并不非常清楚。细胞治疗产品的组织分布和存续时间是影响其有效性和安全性的重要因素,需进行动态观察。目前可选择的示踪技术包括影像学、聚合酶链反应(PCR)和免疫组织化学等技术。PCR技术和免疫组织化学技术的缺点是需要在细胞移植后处死动物,取出相应组织进行检测,存在一定的局限性。随着影像学技术的发展,正电子发射计算机断层成像、单光子发射计算机断层成像、磁共振成像和光学成像等手段为细胞移植后无创和实时评估移植细胞的分布、迁移和归巢提供了新选择。近年来,已有大量关于细胞非临床无创性示踪方法的研究。本综述主要对细胞非临床活体示踪方法的研究进展进行综述,从而为细胞治疗产品的组织分布、迁移和归巢研究提供参考。Cell therapy,as a novel therapy in oncology research and clinical practice,plays an increasingly important role in the treatment of a variety of diseases by repairing damaged tissues or replacing defective cells in the body.However,the mechanisms by which cell therapy products treat diseases and their distribution and migration within the host after cell transplantation are not well understood.The tissue distribution and survival time of cell therapy products are important factors affecting their effectiveness and safety of cell therapy products,and dynamic observations should be made.Currently,alternative technical methods include imaging techniques,polymerase chain reaction techniques,and immunohistochemistry,but the disadvantages of polymerase chain reaction techniques and immunohistochemistry lie in the need to sacrifice the animal after cell transplantation and remove the corresponding tissue for testing,which has some limitations.With the development of imaging techniques,positron emission tomography,single photon emission computed tomography,magnetic resonance imaging,and optical imaging have allowed noninvasive and real-time assessment of the distribution,migration,and homing of transplanted cells after cell transplantation.In recent years,there have been numerous studies on cell preclinical noninvasive tracing methods.This review is a summary of the progress made in preclinical intravital tracing methods of cells,thereby providing reference for tissue distribution,migration and homing studies of cell therapies.
