摘  要:目的使用基因表达谱芯片和miRNA芯片技术分析银屑灵片对寻常型银屑病血瘀证患者治疗前后外周血单核细胞(PBMC)差异表达基因和miRNA,进行关联分析,探讨银屑灵片治疗寻常型银屑病血瘀证的作用机制。方法选择9例寻常型银屑病血瘀证患者,予银屑灵片治疗,并收集健康者9人外周血,制备PBMC,使用基因表达谱芯片和miRNA芯片检测PBMC基因和miRNA表达谱,对结果进行关联分析。结果与健康组比较,治疗前银屑灵片组PBMC的差异miRNA包括hsa-miR-30c-5p等48个miRNA,差异miRNA的靶基因包括PIK3C2B等27个基因;治疗后银屑灵片组PBMC的差异miRNA包括hsa-miR-23c等47个miRNA,差异miRNA的靶基因包括HIVEP2等33个基因。治疗前后银屑灵片组PBMC差异miRNA包括hsa-miR-146b-5p等7个,差异miRNA的靶基因包括MYADM等14个基因。结论通过对治疗前后银屑灵片组患者PBMC表达谱芯片与miRNA芯片结果进行关联分析,提示hsa-miR-146b-5p等7个miRNA及其靶基因DYNLL2等14个基因有可能是银屑灵片治疗寻常型银屑病血瘀证的重要靶点。Objective To analyze the correlation of gene chip and miRNA chip of peripheral blood mononuclear cells of psoriasis vulgaris complicated by blood stasis in patients,aiming to explore its the treatment mechanism using Yin Xie Ling.Methods Totally 9 patients with psoriasis vulgaris complicated by blood stasis and 9 healthy controls were enrolled,Peripheral blood was collected from all subjects,and then the gene expression profile and miRNA expression profile of PBMC were detected to anzy the correlation.Results Compared with the healthy control group,the differential miRNAs of psoriasis patients with blood stasis syndrome before Yin Xie Ling treatment included 48 miRNA involving hsa-miR-30c-5p,and the target genes of differential miRNAs included 27 genes involving PIK3C2B;the differential miRNAs of psoriasis patients with blood stasis syndrome after Yin Xie Ling treatment included 47 miRNAs involving hsa-miR-23c,and the target genes of differential miRNAs involving HIVEP2 included 33 genes;the differential miRNAs in Yin Xie Ling Group before and after treatment included 7 miRNAs involving hsa-miR-146b-5p,and the target genes of differential included 14 genes involving MYADM.Conclusion It is suggested that 7 miRNAs involving hsa-miR-146b-5p and 14 genes involving DYNLL2 may be the important targets for Yin Xie Ling to treat psoriasis vulgaris with blood stasis syndrome through the correlation analysis on the gene profile chip and miRNA chip results of PBMC of psoriasis patients treated with Yin Xie LinG.
