摘 要:目的观察参灵颐肝汤对对乙酰氨基酚(APAP)所致肝损伤小鼠的治疗效果,并探讨作用机制。方法将50只ICR小鼠按照随机数字表法分为空白对照组、模型组、乙酰半胱氨酸组、参灵颐肝汤组、两药联用组,每组10只。空白对照组予0.9%氯化钠注射液0.13 mL/10 g灌胃,其余4组予25 mg/mL APAP溶液0.13 mL/10 g灌胃,每2 d 1次,连续2周,建立急性肝损伤模型。造模后,空白对照组、模型组予0.9%氯化钠注射液0.2 mL/10 g灌胃,乙酰半胱氨酸组予10 mg/mL的乙酰半胱氨酸溶液0.1 mL/10 g腹腔注射,参灵颐肝汤组予3.5 g/mL的参灵颐肝汤0.2 mL/10 g灌胃,两药联用组予3.5 g/mL的参灵颐肝汤0.2 mL/10 g灌胃联合10 mg/mL的乙酰半胱氨酸溶液0.1 mL/10 g腹腔注射,每日1次,连续2周。检测各组小鼠血清天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、总胆红素(TBiL)水平;检测肝脏匀浆液上清白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)、超氧化物歧化酶(SOD)、丙二醛(MDA)水平。取肝脏右叶进行苏木精-伊红(HE)染色,观察肝组织的病理改变情况。结果与空白对照组比较,模型组、乙酰半胱氨酸组小鼠血清AST、ALT、TBiL水平均升高(P<0.05),参灵颐肝汤组小鼠血清AST、ALT水平均升高(P<0.05),两药联用组小鼠血清AST水平升高(P<0.05)。与模型组比较,乙酰半胱氨酸组、两药联用组小鼠血清AST、ALT、TBiL水平均降低(P<0.05),参灵颐肝汤组小鼠血清AST、TBiL水平均降低(P<0.05)。与乙酰半胱氨酸组比较,参灵颐肝汤组、两药联用组小鼠血清AST、TBiL水平均降低(P<0.05)。与参灵颐肝汤组比较,两药联用组小鼠血清ALT水平降低(P<0.05)。与空白对照组比较,模型组、乙酰半胱氨酸组小鼠肝组织IL-6、MDA水平均升高(P<0.05),SOD水平均降低(P<0.05);参灵颐肝汤组、两药联用组小鼠肝组织SOD水平均降低(P<0.05)。与模型组比较,乙酰半胱氨酸组小鼠肝组织IL-6、TNF-α、MDA水�Objective To observe the efficacy and mechanism of Shenling Yigan Decoction(SLYGD)for rat due to acetaminophen(APAP)induced liver injury.Methods Fifty ICR rats were randomly divided into blank group and four experimental groups,10 cases in each;except for blank group was given 0.9%sodium chloride injection by gastric irrigation with 0.13 mL/10 g,the rats in experimental groups were administered with 25 mg/mL APAP solution by gastric irrigation with 0.13 mL/10 g,once every 2 days,to induce liver injury model.Subsequently,the model rats were divided into a model group[Gavaged 0.9%sodium chloride injection,0.2 mL/10(g·d)],N-acetylcysteine(NAC)group[injected,10 mg/mL AC solution,0.1 mL/10(g·d)],SLYGD group[Gavaged 3.5 g/mL 0.9%sodium chloride injection,0.2 mL/10(g·d)],combined group(the combination therapy of NAC group and SLYGD group),blank control group(similarly treated as modle group).The rats in each group were dosed once a day,for consecutive 2 weeks.The key observation was aspartate aminotransferase(AST),alanine aminotransferase(ALT),total bilirubin(TBiL),interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),superoxide dismutase(SOD)and malondialdehyde(MDA)in groups,the right lobe of liver was stained with hematoxylin-eosin(H&E)to observe the pathological changes.Results Compared with blank group,model group and NAC group exhibited significantly increased expression of AST,ALT,TBiL in serum(P<0.05),up-regulated IL-6,MDA expression(P<0.05),reduced SOD in liver tissue(P<0.05);SLYGD group showed elevated expression of AST,ALT in serum(P<0.05),decreased SOD in liver tissue(P<0.05);combined group displayed elevated expression of AST in serum(P<0.05).Compared with model group,NAC group and combined group exhibited significantly decreased expression of AST,ALT,TBiL in serum(P<0.05);NAC group exhibited significantly decreased expression of IL-6,TNF-αand MDA in serum(P<0.05),increased SOD in liver tissue(P<0.05);SLYGD group showed decreased expression of AST,TBiL in serum(P<0.05),SLYGD group and combined group exhib
