Effect of Fried Semen Ziziphi Spinosae on Dynamic Changes of Neuronal Information Coding in Basolateral Amygdala of Anxiety Rats

摘  要:目的探讨炒酸枣仁对焦虑大鼠基底外侧杏仁核(BLA)神经元信息编码的动态变化的干预作用。方法将SD雄性大鼠随机分为模型对照组与炒酸枣仁组,灌胃给予炒酸枣仁组大鼠炒酸枣仁水煎液(17.5 g·kg-1),模型对照组给予同等体积纯化水,每日1次,连续7 d。每日给药0.5 h后采用旷场实验方法和在体多通道神经同步记录技术,监测两组大鼠的行为学及BLA区神经元信息编码。结果随测试时间的增加,大鼠的探索行为逐渐减少,焦虑样行为逐渐减少,BLA区神经元动作电位放电频率在1~2 d上升,2~7 d持续下降。与模型对照组比较,炒酸枣仁组大鼠进入中央格次数在第1天明显增加(P<0.01);中央格运动时间在第1~3,6天明显增加(P<0.05或P<0.01);中央格/总运动时间(%)在第1,3,5~7天明显增加(P<0.05或P<0.01);BLA区NeuronⅠ的动作电位放电频率增加值在第1,3~6天明显减少(P<0.05或P<0.01);NeuronⅡ的动作电位放电频率增加值在第1,3,4,6,7天明显减少(P<0.05或P<0.01)。结论重复暴露于旷场环境中大鼠建立习惯化学习,表现为运动性逐渐降低、焦虑水平逐渐下降、BLA区神经元动作电位放电频率先增加后下降。炒酸枣仁能明显降低大鼠的焦虑样行为,具有一定的抗焦虑作用,其作用机制与抑制BLA区神经元动作电位的过度放电、改变BLA区神经元信息编码有关。Objective To investigate the effect of fried Semen Ziziphi Spinosae on the dynamic changes of neuronal information coding in basolateral amygdala(BLA)neurons of anxiety rats.Methods Male SD rats were randomly divided into model control group and Semen Ziziphi Spinosae group.Rats in fried Semen Ziziphi Spinosae group were given decoction of fried Semen Ziziphi Spinosae(17.5 g·kg-1)by gavage,and model control group was given the same volume of distilled water once a day for 7 consecutive days.After 0.5 hours of daily administration,open field test and in vivo multi-channel nerve synchronous recording technology were used to monitor the behavior and neuronal information coding in BLA region in these two groups.Results With the increasing of testing days,the exploratory behavior and anxiety-like behavior of rats gradually decreased,and the spike firing rate of BLA neurons increased in 1-2 d and decreased continuously in 2-7 d.Compared with the model control group,the number of rats that entered the central grid in fried Semen Ziziphi Spinosae group was significantly increased at the 1st day(P<0.01);the central grid exercise time was significantly increased on the 1st-3rd and 6th days(P<0.05 or P<0.01);the central grid/total exercise time(%)was significantly increased on the 1st day(P<0.05 or P<0.01);the increase value of spike firing rate of NeuronⅠin the BLA region was significantly decreased on the 1st,and at 3-6th days(P<0.05 or P<0.01);and the increase value of spike firing rate of neuronⅡwas significantly reduced on the 1st,3rd,4th,6th and 7th days(P<0.05 or P<0.01).Conclusion Rats exposed to open-field environment repeatedly established habitual learning,which showed that motor activity decreased gradually,and anxiety level decreased gradually,and spike firing rate of BLA region increased at the beginning and then decreased.Fried Semen Ziziphi Spinosae could significantly reduce the anxiety-like behavior of rats,and had anti-anxiety effect,and the mechanism of anti-anxiety effect is related to the inhibiti
