摘　　要：目的建立高效液相色谱(HPLC)波长切换法同时测定"银板方"中(R,S)-告依春、绿原酸、芍药内酯苷、连翘酯苷A和次野鸢尾黄素含量的方法。方法采用Agilent InfinityLab Poroshell 120 SB-C18(3.0 mm×150 mm,2.7μm)色谱柱,0.1%磷酸乙腈-0.1%磷酸水为流动相,梯度洗脱,流速为0.8 mL/min,柱温35℃;波长切换方式0~4 min为240 nm,4~6.5 min为325 nm,6.5~9 min为230 nm,9~21 min为328 nm,21~24 min为265 nm。结果(R,S)-告依春、绿原酸、芍药内酯苷、连翘酯苷A、次野鸢尾黄素分别在0.145~18.560(r=1)、3.566~228.200(r=1)、2.281~292.000(r=1)、6.312~808.000(r=1)、0.238~30.480(r=1)g范围内与峰面积线性关系良好,平均回收率分别为97.86%、102.09%、99.06%、98.63%、101.51%,RSD分别为1.90%、1.43%、1.62%、1.40%、1.94%(n=9)。结论HPLC波长切换法准确、可靠、重复性好,可用于"银板方"的质量评价。Objective To develop an HPLC wavelength switching method for determining the contents of(R,S)-goitrin,chlorogenic acid,albiflorin,forsythoside A,irisflorentin in Yinban Prescription.Methods The separation was performed on an Agilent InfinityLab Poroshell 120 SB-C18(3.0 mm×150 mm,2.7μm)at 35℃using a gradient elution with 0.1%phosphoric acid acetonitrile and 0.1%phosphoric acid water.The flow rate was 0.8 mL/min and the detection wavelength was set at 240 nm within 0-4 min,325 nm within 4-6.5 min,230 nm within 6.5-9 min,328 nm within 9-21 min,265 nm within 21-24 min.Results(R,S)-goitrin,chlorogenic acid,albiflorin,forsythoside A,risflorentin had a good linear relationship with the peak area in the range of 0.145-18.560(r=1),3.566-228.200(r=1),2.281-292.000(r=1),6.312-808.000(r=1),0.238-30.480(r=1)g,respectively.The average recoveries were 97.86%,102.09%,99.06%,98.63%,101.51%,with RSD 1.90%,1.43%,1.62%,1.40%,1.94%(n=9),respectively.Conclusion The HPLC wavelength switching method is accurate,reliable and reproducible for quality evaluation of Yinban Prescription.
