摘  要:目的探讨miR-1193在宫颈癌组织的表达及其对宫颈癌(CC)细胞增殖和迁移的影响。方法收集我院接受手术治疗的宫颈组织60例,取宫颈组织[包括高级别宫颈上皮内瘤变(HSIL),低级别宫颈上皮内瘤变(LSIL)和宫颈癌(CC)]及病灶旁正常组织。用逆转录实时荧光定量聚合酶链反应(qRT-PCR)检测miR-1193表达水平,并分析其表达水平与临床特征的相关性。在宫颈癌细胞株(HeLa、SiHa)中用脂质体转染法转染miR-1193和miR-C,用细胞计数(CCK-8)法、Transwell实验和细胞划痕实验检测miR-1193对宫颈癌细胞株(HeLa、SiHa)的增殖、侵袭和迁移能力的影响。结果同正常组织对比,miR-1193在LSIL,HSIL以及CC组织中呈现低表达,且其表达量逐渐降低(P<0.01)。其表达水平与高危型人乳头瘤病毒(HPV)感染有关(P<0.05),但与患者年龄、肿瘤大小、脉管癌栓等均无明显相关性。过表达miR-1193可以抑制CC细胞株(HeLa、SiHa)的增殖、侵袭和转移(P<0.01)。结论 miR-1193在CC组织表达降低,上调miR-1193表达后可以抑制CC细胞株(HeLa、SiHa)的增殖、侵袭和转移。Objective To investigate the expression of miR-1193 in cervical tissue and its effect on proliferation and migration in cervical cancer(CC)cells. Methods Sixty cases of cervical tissue [including high-grade cervical intraepithelial neoplasia(HSIL), low-grade cervical intraepithelial neoplasia(LSIL), cervical cancer(CC)]and normal tissues adjacent to the lesions were collected. The expression of miR-1193 was detected by real-time quantitative polymerase chain reaction(qRT-PCR), and the correlation between the expression level and clinical features was analyzed. miR-1193 and miR-C were transfected into CC cells(HeLa, SiHa) by lipofectamine transfection. CCK-8, Transwell test and cell scratch test were used to detect the effects of miR-1193 on the proliferation, invasion and migration of cervical cancer cell lines(HeLa, SiHa). Results Compared with normal tissues, miR-1193 was low expressed in LSIL, HSIL and CC tissues, and its expression level gradually decreased(P<0.01). The expression level was associated with high-risk human papilloma virus(HPV) infection(P<0.05), but not with age, tumor size, vascular tumor thrombus( P > 0. 05). Over expression of miR-1193 inhibited the proliferation,invasion and metastasis of CC cell lines( P < 0. 01). Conclusion The expression of miR-1193 in CC was decreased. Up-regulation of miR-1193 can inhibit the proliferation,invasion and metastasis of CC cell lines( He La,Si Ha).
