摘  要:目的研究桔梗汤不同部位基于IL-17A/TNF-a信号通路对COPD模型大鼠的影响,初步筛选桔梗汤治疗COPD的有效部位。方法建立COPD大鼠模型,将40只SD大鼠分为生理盐水组、模型组、桔梗汤总皂苷组、桔梗汤总多糖组和桔梗汤总黄酮组,经过28天的治疗后,观察分析大鼠一般情况及体重变化、大鼠肺组织病理学改变及气管壁厚度变化、大鼠血清中IL-17A、TNF-a、IFN-α和IFN-β的表达。结果桔梗汤总皂苷和总黄酮对大鼠体重变化、大鼠气管壁厚度变化、大鼠血清中IL-17A、TNF-a、IFN-α和IFN-β的表达和模型组比较有显著性差异(P<0.05),桔梗汤总多糖对上述指标和模型组比较无显著性差异(P>0.05)。结论桔梗汤总皂苷和总黄酮可通过IL-17A/TNF-a信号通路显著改善COPD大鼠肺功能,并初步确定桔梗汤防治COPD的有效部位为总皂苷和总黄酮。Objective To investigate the effect of different components in Jiegeng Decoction on COPD rats based on IL-17 A/TNF-a signaling pathway,and to screen the effective components in Jiegeng Decoction for COPD.Methods Totally 40 healthy rats were selected and randomly divided into 4 groups,including saline treated group,COPD group,total saponins in Jiegeng Decoction,total polysaccharides in Jiegeng Decoction and total flavonoids in Jiegeng Decoction.Body weight change,histochemistry on lung tissue,thickness change of tracheal wall,serum IL-17 A,TNF-α,IFN-αand IFN-βlevel were checked after a 28-day treatment.Results Treatments of total saponins and total flavonoids significantly affected rat body weight,thickness of tracheal wall,serum IL-17 A,TNF-α,IFN-αand IFN-βlevel compared with COPD group(P<0.05).In contrast,total polysaccharides treatment had undetectable effect compared with COPD rats regarding the above measurements(P>0.05).Conclusion Total saponins and total flavonoids in Jiegeng Decoction can significantly improve rat lung functions in terms of IL-17 A/TNF-αsignaling pathways.Therefore,we conclude that the effective components in Jiegeng Decoction for the relief of COPD rats are total saponins and total flavonoids.
