摘 要:为了解EM菌对中华绒螯蟹(Eriocheir sinensis)免疫力的影响,本研究以中华绒螯蟹为研究对象,在成蟹养殖过程中定期使用EM菌,探究了水体中添加EM菌对中华绒螯蟹抗氧化酶系统和非特异性免疫的影响。结果显示:EM菌能提高特定发育阶段河蟹血清中过氧化氢酶(CAT)活力,降低丙二醛(MDA)的含量,增强河蟹抵抗氧化应激的能力。半颗粒细胞(SGC)是中华绒螯蟹血细胞的主要类群,约占51.50%~58.08%;EM菌诱导下,河蟹SGC细胞升高2.97个百分点,但与对照组之间差异不显著。EM菌能够诱导上调河蟹肝胰腺免疫相关基因Toll2、Tube、Hsp90、Dorsal表达,一定程度上能提高机体对异物的识别能力和防御应激损伤的能力;EM菌组血清谷丙转氨酶(ALT)的水平较低,表明器官损伤较低;同时较高的碱性磷酸酶(ALP)活性意味着免疫解毒能力的提升。综上,水体中添加EM菌有助于提高中华绒螯蟹抵抗氧化应激的能力,也可通过调控河蟹免疫基因的表达和相关酶活性,增强其非特异性免疫力。The Eriocheir sinensis,is an important economic and widely cultivatied aquaculture species in China.The present study was conducted to determine the effect of EM on the antioxidant capacity and immune ability in E.sinensis by adding EM to water during the process of adult E.sinensis culture.The results indicated that EM could increase the activity of catalase(CAT)in the serum at specific developmental stages,reduce the content of malondialdehyde(MDA),and enhance the ability of crabs to resist oxidative stress.Semigranular cells(SGC)are the main type of hemocytes in E.sinensis,accounting for 51.50%~58.08%.Under the induction of EM,the proportion of SGC cells in E.sinensis increased by 2.97 pexcentage points,albeit there was no significant difference compared to the controls.EM can up-regulate the mRNA levels of immune-related genes,such as Toll2,Tube,Hsp90,and Dorsal in the hepatopancreas of E.sinensis.Through this EM could improve the ability in recognition of xenobiotics and the defense ability against stress damage.The serum alanine aminotransferase(ALT)level in EM group was lower,reflective of a lesser degree of hepatopancreas damage.Meanwhile,the higher alkaline phosphatase(ALP)activity means the improved immune detoxification ability.In conclusion,adding EM in water can improve the ability of E.sinensis to resist oxidative stress,and enhance its non-specific immunity by regulating the expression of immune genes and related enzyme activities.
