摘  要:为提高蒸汽闪爆处理后牛角蛋白酶解率和探究在不同溶剂中溶解度的条件,试验选取牛角粉(牦牛角)为原料,溶解度和酶解率为评价指标,在单因素试验基础上,采用响应面三因素三水平中心组和试验对酶解条件进行优化,最佳酶解工艺:酶添加量1.4%、底物浓度为6%、时间8.0 h、温度为60℃、pH=8.在此蒸汽闪爆处理酶解条件下压力4.0 MPa时溶解度最好,牛角蛋白酶解率达到21.07%.牛角蛋白在尿素溶液中的溶解率达到60.7%,在盐酸胍溶液中的溶解率为53.01%.结果表明,该酶解工艺合理可行,其牛角蛋白蒸汽闪爆处理后溶解度明显提高,具有较好的酶解效果.本研究可为屠宰副产物进行高效、经济、环保处理提供数据参考.In order to explore the conditions for improving the proteolysis rate of horns and the solubility in the different solvents after steam flash explosion treatment, the horn powder(yak horn) was selected as the raw material in the present experiment.Tthe solubility and enzymatic hydrolysis rate were evaluated.Based on the single factor experiment, the response was adopted.The three-factor three-level center group and experiment were used to optimize the enzymatic hydrolysis conditions.The best enzymatic hydrolysis process: enzyme addition amount 1.4%,substrate concentration 6%,time 8.0 h, temperature 60 ℃ and 8 of pH.Under the steam flash explosion treatment enzymatic hydrolysis conditions, the solubility is the best at 4.0 MPa, and the proteolysis rate of cow keratin was 21.07%.The solubility rate of cow keratin in urea solution was 60.7%.It was dissolved in the guanidine hydrochloride solution.The rate was 53.01%.The results showed that the enzymatic hydrolysis process was reasonable and feasible.The solubility of bovine keratin was obviously improved after steam flash explosion treatment.It had a better enzymatic hydrolysis effect.It showed that the above results can be used as a data reference for more efficient, economical and environmentally friendly treatment of slaughter by-products.
