摘  要:【目的】拟通过蝉花野外繁育菌株的子实体培养筛选和分子标记分析,建立蝉花野外繁育菌株鉴别体系,获得可用于生产的优质菌株。【方法】以蝉花A17-7-1菌株野外繁育获得的7个蝉花菌株进行子实体培养筛选,并用A17-7-1菌株特异的SCAR分子标记对菌株进行分析。经SCAR标记确认的繁育菌株再以ISSR分子标记分析判定繁育菌株遗传性状。【结果】经子实体培养筛选,2个菌株子实体产量高于A17-7-1;SCAR分子标记分析显示,从这2个子实体产量提高的菌株基因组NDA中扩增出A17-7-1菌株特有的DNA片段;ISSR分子标记分析表明,一株子实体产量明显提高的菌株与A17-7-1具有完全相同的条带,另一个子实体产量提高不显著的菌株有2个条带缺失。【结论】野外繁育蝉花可提高子实体产量,SCAR标记可快速鉴别菌株是否为繁育菌株,ISSR标记能够初步判断菌株遗传特性。【Objective】The present paper aimed to establish the identification system of Isaria cicadae field breeding strains through the fruit body culture screening and molecular marker analysis and obtain high-quality strains that could be used for production.【Method】Seven Isaria cicadae isolates by breeding in bamboo forest with strain A17-7-1 were screened for fruit body cultivation,and the isolates were analyzed with the specific SCAR molecular marker of strain A17-7-1.The breeding strains confirmed by the SCAR marker were then analyzed by ISSR molecular markers to determine the genetic characteristics.【Result】The fruit body yield of two strains was higher than that of A17-7-1;SCAR molecular marker analysis showed that the two strains amplified DNA fragments unique to the A17-7-1 strain;ISSR molecular marker analysis showed that one strain with significantly increased fruit body yield had exactly the same bands as A17-7-1,and the other strain with insignificant increase in fruit body yield had 2 bands missing.【Conclusion】Breeding Isaria cicadae in bamboo forest could increase the yield of fruit bodies,SCAR marker could quickly identify whether the isolates were breeding strains,and ISSR marker could preliminarily judge genetic characteristics of the strains.
