摘　　要：目的对SD大鼠垂体自发性病变Rathke裂囊肿发生情况进行观察分析,获得安全性评价试验中SD大鼠的背景病变资料。方法收集近2年完成的10项90天经口毒性试验中的对照组及高剂量组动物,共计400只17~19周龄SD大鼠,雌雄性别各半,对各只动物的垂体病理切片在光学显微镜下进行组织学观察,对Rathke裂囊肿进行诊断并统计其发生率。结果在总计400只SD大鼠中,共报告10例Rathke裂囊肿,总体发生率为2.5%。在200只雌性SD大鼠中报告4例,发生率2%;在200只雄性SD大鼠中报告6例,发生率3%。结论在17~19周龄SD大鼠中,垂体Rathke裂囊肿的发生率约为2.5%,未表现出性别差异。Objective To investigate and analyze the incidence of spontaneous pituitary Rathke′s cleft cysts in Sprague-Dawley(SD)rats,and to accumulate background lesions data of SD rats for safety evaluation.Methods Four hundreds SD rats(17-19 weeks old,half in each gender)were recruited from control and high dose groups in 10 projects of 90-day oral toxicity test completed in last two years.Histological observation of pathological changes was conducted in pituitary gland of each animal.Diagnosis of Rathke′s cleft cysts was isolated and counted its incidence.Results In a total of 400 SD rats,10 cases of Rathke′s cleft cyst were reported,and the overall incidence was 2.5%.Four cases were reported in 200 female SD rats with an incidence rate of 2%,and six cases were reported in 200 male SD rats with an incidence of 3%.Conclusion In SD rats aged 17-19 weeks,the incidence of pituitary Rathke′s cleft cyst was 2.5%,showing no gender difference.
